桑树染色体制片的直接酸解去壁法及其实用性研究

摘　　要：本文研究出桑树染色体制片的直接酸解去壁法：直接取桑嫩芽幼叶投入“酸醇去壁固定液”（浓盐酸∶４５％醋酸∶纯乙醇＝１∶０．５∶０．５容积比）中３～５ｍｉｎ，然后将材料水洗几次浸入水中后低渗１０ｍｉｎ，再经染色即制备成染色体标本。验证了在保证染色体制片观察效果前提下，样本材料采集后在室温和冰箱保存的时间。并与其它制片方法从制片效果、工作效率、成本等方面进行比较。从而证明本方法具有操作简单、省时省力、成本低、效率高、有较大的实际应用价值。a method for chromosome preparation in mulberry by the cell wall being digested direct with acid method was developed . The buds or tender leaves of mulberry were directly put into the fluid ( 10 ml the thick hydrochloric acid + 5 ml 45% acetic acid + 5 ml 100% alcohol) for 35 min. Then they were took out and washed several times with water, and put into water for 10 min for back lowosmosis, then were stained to prepare the chromosome specimen. the sample of mulberry could be stored in both the roomtemperature and 712 in the refrigerator for hours to 36 hours before used, with the some result as fresh samples .It was also compared to other methods of preparation specimen in effect, efficiency and cost of preparation ,and showed it was simper and effective method.
