摘 要:目的 探讨中药牛膝来源的多糖刺激粒细胞集落形成的能力。方法 使用软琼脂培养体系,培养骨髓造血干细胞,设置粒细胞集落刺激因子(G-CSF)、牛膝多糖(APP)、G-CSF和APP联合作用实验组以及对照组(0.1ng/ml G-CSF),分别检测有效集落数。结果 G-CSF联合APP能有效促进粒系集落形成单位(CFU-G)的形成,与G-CSF单独作用相比差异显著(P<0.05)。0.8mg/ml APP对CFU-G的刺激形成作用与1ng/ml的G-CSF相似。结论 APP具有刺激粒细胞集落形成的能力,并与G-CSF有明显的协同作用,有望成为一种有效的粒细胞刺激因子。Objective To explore the capability of polysaccharide derived from a Chinese herbal medicine, achyranthes - piaentata polysaccharide (APP), to stimulate granulocyte clone formation. Methods The semi - solid agarose system was used to culture stem cells derived from bone marrow. A set of experiment and control were established to check the effective clone formation stimulated with G - CSF and/or APP. Results When there exist G - CSF in bone marrow cell culture, introducing APP could promote effectively the number of G - CFU (P < 0. 05, compared with the G - CSF only control). The effects of 0. 8mg/ml APP to stimulate granulocyte system stem cells was near or equal to lng/ml G - CSF. Conclusion APP can stimulate granulocyte clone formation; and has synergy effect with G - CSF. It is hopeful that APP can be used as an effective granulocyte stimulating factor.
