摘 要:目的 观察外源性褪黑激素 (MT)对睾丸和附睾一氧化氮合酶 (NOS)的影响。方法 HE染色 ,免疫组化SABC法测定三种NOS的分布情况。结果 三种NOS主要分布于Leydig细胞 ,其中NOS1阳性细胞最多 ,2 0~ 30d处理组的阳性细胞数总体多于 30~ 4 0d处理组 ,但呈被MT抑制的趋势 ;30~ 4 0d处理组则相反。在MT注射组中附睾上皮出现三种NOS阳性细胞 ,间质微血管内皮出现NOS3阳性细胞。少数精母细胞呈NOS阳性。结论 褪黑激素性成熟前抑制NOS在Leydig细胞中的功能表达 ,性成熟期则刺激NOS在睾丸和附睾中的功能表达 ;NOS与雄性激素的分泌和精子功能的成熟有关。Objective To observe the melatonin influence to the distribution of NOS in testis and epididymis. Methods HE stain, immunohistochemistry SABC to determine the change of three kinds of NOS. Results Three kinds of NOS spreads mainly in leydig cell, in which NOS1 positive cell with a maximum of, positive cells in the handling grow of 20~30 d is overall more than the handling group of 30~40 d and show the tendency that inhibited by MT. The handling group of 30~40 d is opposite to he handling grow of 20~30 d. In groups of injecting MT, three kinds of NOS positive cell appearance in epididymal epithelium, NOS3 positive cell appearance in microvessel endothelium of interstitium. Few NOS positive cells appearance in spermatogenic cells. Conclusions Exo-melatonin inhibits the activity of NOS in leydig cell before sexual maturity and stimulates the activity of NOS in testis and epididymis in the period of sexual maturity. NOS is related to secretion of androgen and maturation of sperm function.
