摘  要:目的:探讨地塞米松对内毒素诱导的大鼠星形胶质细胞中诱导型一氧化氮合酶(iNOS)活性的影响。方法:应用Griess法,动态测定内毒素(LPS)诱导体外培养的大鼠星形胶质细胞iNOS活性的变化以及地塞米松(DEX)对其的影响。结果:细菌LPS能够刺激星形胶质细胞iNOS活性的表达,且与LPS剂量和作用时间有关,DEX对星形胶质细胞iNOS活性表达具有负调节作用,而且与加入DEX的间隔时间有关。结论:星形胶质细胞iNOS可能参与中枢神经系统细菌感染的病理过程,而DEX能抑制这种损害作用。Objective:To examine the effect of dexamethasone on the inducible nitric oxide synthase(iNOS) activity induced by endotoxin (LPS) in cultured rat astrocytes.Methods:INOS activity in normal astrocytes,LPS induced astrocytes,and dexamethasone treated LPS induced astrocytes was detected by Griess methods.Results:INOS activity was increased by endotoxin inducement in a dose and time dependent manner.Dexamethasone inhibited iNOS activity in astrocytes induced by endotoxin,and this inhibition was associated with the delay of dexamethasone addition after LPS stimulation.Conclusion:It was suggested that the iNOS of astrocytes might have a part in the pathology of bacterial infection in central nervous system,and it's role could be decreased by DEX.
