摘 要:从抗人膀胱癌改形噬菌体抗体库中筛选出一组相对亲和力常数和细胞结合活性相近的单链抗体 ,而它们的结合活性均明显高于起始的鼠源单链抗体。在亲和力淘选的过程中 ,这一组抗体的表达量存在显著差异 ,这在一定程度上影响到筛选过程中ELISA鉴定的结果 ,使整个筛选过程并不仅仅是对高亲和力抗体的筛选 ,也同时得到了高表达量的抗体。In our work to select high affinity antibodies from reshaped phage antibody library against human bladder carcinoma, we found a panel of antibodies with much different antigen binding activity in library screening had similar relative affinity constants. The result of cytometry also indicated that these antibodies had similar cell binding activity but much higher than that of mouse counterpart. And we found that the expression level of these antibodies was much different during the course of biopanning , which partly accounted for the much difference in ELISA. It also indicated this selection was less than straightforward for antibody affinity due to difference in phage antibody expression. But we can use the method to select antibodies with increased expression level.
