摘 要:目的 :建立一种一阶导数紫外分光光度法测定水飞蓟宾脂质体包封率及脂质体中水飞蓟宾含量。方法 :水飞蓟宾脂质体溶液经SephadexG10 0 柱分离后 ,以水飞蓟宾一阶导数紫外扫描图谱在 2 77nm和 2 94nm处出现的特征峰和特征谷的振幅ΔA进行定量。结果 :方法的线性范围为 0 .5~ 3 0 .0mg·L- 1(r =0 .9998) ,回收率为10 1.7% ,精密度为 1.8%。采用该法测定药脂比为 0 .0 2~ 0 .0 6的水飞蓟宾脂质体 ,包封率为 65.1%~ 83 .0 % ,脂质体中水飞蓟宾平均含量为 760 .4mg·L- 1(n =4)。结论 :一阶导数分光光度法准确、简便 。Objective: To determine the encapsulation efficiency of silybin lipos omes. Method: After Silybin liposomes solution was separat ed through sphadex G- 100 column, eluent collected was determined by the first derivative UV spectrome try. Result: The calibration curve of Silybin was linear i n the range of 0.5~30 .0 mg·L -1 (r=0.999 8).The average recovery was 101.7%. The relat ive standard de viation was 1.8%. The results showed that the encapsulation efficiency ranged fr om 65.1%~83.0% when the drug /phospholipid ratio varied from 0.02~0.06. The co ncentration of SLB in liposomes prepared by optimum technique was 760.4 mg·L -1 ( n=4). Conclusion: The method is accurate, simple and s uitable for the quality control of Silybin liposomes.
