救脑益智胶囊水提物对人神经母细胞瘤株SY5Y信号转导的影响

摘　　要：目的 :通过观察救脑益智胶囊对人神经母细胞瘤株SY5Y信号转导的影响 ,探讨此药促进神经细胞生长的可能机理。方法 :以人神经母细胞瘤株SY5Y为细胞模型 ,分为正常对照组和加药组。收集对照组及加药组细胞裂解物 ,取上清用Lowry法进行蛋白定量 ,将总蛋白量相等的样品进行免疫沉淀Westernblot分析。 结果 :与正常对照组比较 ,加药组SY5Y细胞蛋白激酶B(proteinkinaseB ,Akt/PKB) ,cAMP反应元件结合蛋白 (cAMPresponseelementbindingprotein ,CREB)、磷酸化的CREB (phosphorylatedCREB ,P CREB)表达增加 ,细胞色素C(cytochromeC ,cytC1)表达减少。结论 :救脑益智胶囊的生药水提取液能促进SY5Y神经母细胞存活信号通路中Akt/PKB ,P CREB的表达 。Objective: To observe the effect of Jiunao yi zhi capsul on the signal transduction pathway of SY5Y cell lines and explore the mechanism of the function of the capsul′s enhancing neuronal growth. Method: Human neuroblastoma was used as cell models and they were d ivided into control group and experimental group. Supernatant of cell lysate was taken and immunop recipitation was done with antibodies to proteins related to signal transduction pathway, and the immunoprecipitates were analyzed by Western blotting. Result: After treatment with Jiunaoyizhi capsul, expression of Akt /PKB, CREB, P-CREB was clearly increased and expression of cytochrome C decreas ed more than the control group. Conclusion: Jiunaoyizhi ca psul can promote expression of some proteins related with signal transduction pathwa y in SY5Y cell lines. Mechanism of Jiunaoyizhi capsul′s enhancin g neuronal growth is relevant to expression of some proteins in signal transduct ion pathway.
