摘  要:目的观察黄芩苷、栀子苷配伍组分(BG)对脂多糖(LPS)诱导小鼠小胶质细胞激活损伤的影响。方法体外培养BV2小鼠小胶质细胞株,取生长良好的细胞分别加入LPS(终浓度500ng/m L)和不同浓度黄芩苷、栀子苷配伍组分(终浓度12.5、25、50μg/m L)进行干预,并设置正常对照组;采用MTT法检测脂多糖对BV2细胞存活率的影响;取培养24 h细胞上清液,用Griess法检测NO生成量。结果 (1)MTT检测结果显示,与对照组相比,LPS单独刺激的BV2小胶质细胞活性明显降低,有统计学差异;与LPS单独处理的模型组细胞相比,给药组(LPS+BG)有效的提高了LPS处理后的BV2小胶质细胞活性,有统计学意义。(2)Griess法检测结果显示:与对照组相比,LPS可明显促进BV2细胞NO的生成,有统计学意义;加入黄芩苷栀子苷配伍组分可抑制LPS诱导的BV2小胶质细胞NO的生成,与LPS组差异有统计学意义。结论黄芩苷、栀子苷配伍组分可显著提高脂多糖诱导的BV2小胶质细胞的存活率,同时显著降低脂多糖诱导BV2细胞NO的表达,初步判断黄芩苷、栀子苷配伍组分可抑制脂多糖所致BV2小胶质细胞的激活与损伤,具有一定的神经保护作用。Objective This report aimed to explore the synergistic effect of Baicalin and Geniposide(BG) on BV2 cell activation damage caused by lipopolysaccharide(LPS). Methods Cultured of BV2 murine microglial cell line in vitro, BV2 cells were added to LPS(final concentration 500ng/m L) and different concentrations of Baicalin and Geniposide(BG)(final concentration 12.5,25,50μg/m L) intervention, and set up the normal control group. MTT Methods were used to compare the difference in BV2 cells between the control group and BG treated group. The accumulated nitrite in the culture supernatants was measured using the Griess reaction method. Results(1) the MTT test showed that LPS-stimulated BV2 cells' viability was significantly decreased compared with the contral group; compared with LPS alone treated cells,drug group(LPS+BG) effectively improves the LPS-stimulated BV2 cells' viability.(2) The accumulated nitrite in the culture supernatants was measured using the Griess reaction method that: compared with the control group, LPS can obviously promote the BV2 cells' NO generation; while the drug group(LPS+BG) can effectively inhibit the generation of NO whitch stimulated by LPS. Conclusion Baicalin and Geniposide(BG) compatibility group points can significantly improve survival rate of LPS-stimulated BV2 cells, also significantly reduced the LPS induced expression of NO.These findings suggest that synergistic drug combination of Baicalin and Geniposide can inhibit LPS induced BV2 microglial cells activation and injury, and have a neuroprotective effect.
