摘  要:目的通过实验观察股骨头局部缺氧诱导因子-1ɑ(hypoxia inducible factor-1ɑ,HIF-1ɑ)在正常与缺血股骨头坏死骨组织中的表达差异,探讨HIF-1ɑ在股骨头坏死中的发病相关机制。方法取正常股骨头、缺血坏死性股骨头骨组织各12例,样本组织2-4g用于HIF-1ɑ基因的RT-PCR。结果 HIF-1ɑ表达量在正常股骨头骨组织中与坏死组织相较更少,正常股骨头骨组织中RT-PCR的2^(-ΔΔCt)平均值为0.46±0.22,而在坏死股骨头骨组织中为0.55±0.24,两者比较差异存在统计学意义(P<0.05)。结论正常股骨头骨组织中的HIF-1ɑ基因与缺血坏死性股骨头骨组织具有统计学差异,故缺血缺氧导致股骨头骨组织中HIF-1ɑ基因表达量的增多可能为股骨头缺血坏死的重要原因。Objective Through the experimental observation of femoral head local hypoxia inducing factor-1ɑ(hypoxia inducible factor-1ɑ,HIF-1ɑ) in normal and ischemic necrotic femoral head expression differences in bone tissue, to explore HIF-1ɑ in the pathogenesis of femoral head necrosis. Methods Taking each normal, ischemic necrotic femoral head bone 12 cases, femoral head sample group 2-4 g for HIF-1ɑ gene RT-PCR. Results HIF-1ɑ expression was less in normal bone tissue than in necrotic tissue, the mean value of RT-PCR 2-ΔΔCt in normal bone tissue was 0.46±0.22, in necrotic bone tissue, it was 0.55±0.24, the difference was statistical significance between the both(P< 0.05).Conclusion HIF-1ɑ gene in normal and ischemic necrotic femoral head bone tissue was statistically significant, and as a result of ischemia hypoxia HIF-1ɑ gene expression increased quantity is probably one of the important reasons of the ischemic necrosis of the femoral head.
