胶原在胎兔、成兔正常发育及创伤愈合过程中的变化

摘  要:目的 :为了探索胎兔及成兔在正常发育及创伤愈合过程中胶原合成特点 ,分析胎儿型及成年型创伤愈合差别的原因 .方法 :用胃蛋白酶对胎兔及成兔皮肤的胶原进行提取 ,用酸水解法使胶原水解 ,采用氯胺T法进行羟脯氨酸 (HP)测定 .结果 :①正常成兔的HP含量明显高于正常胎兔 ,而正常胎兔不同孕期之间却无差别 .②胎兔皮肤创伤后 (1~ 6d)与对应的正常胎兔相比 ,只表现出第 1dHP显著增高 .③成兔皮肤创伤后与正常成兔相比 ,除第 1日无差别外 ,后 5dHP显著增高 .④胎兔皮肤创伤后HP动态变化呈现线性下降趋势 ,而成兔呈现线性上升趋势 .结论 :成兔皮肤合成胶原的能力高于胎兔皮肤 ;创伤后 ,胎兔及成兔均表现出胶原合成增加 ,而前者持续短暂 ,预示着愈合迅速 ;而后者持续增高 。AIM: To study the character of collagen synthesis of fetal and adult skin in normal development and wound healing and to explore the reason for the difference between fetal type and adult type in wound healing. METHODS: Collagen in fetal and adult rabbit skin was extracted with trypsin and hydrolyzed with acid hydrolysis, and hydroxyproline (HP) was determined by chloramines T method. RESULTS: ① HP contents in normal adult rabbits were much higher than those in normal fatal rabbits but with no difference between those of fetal rabbits at different pregnant time; ② Compared with those in normal fetal rabbits, HP contents at first day after injury elevated obviously in fetal rabbits; ③ Compared with those in normal adult rabbits, HP contents in adult rabbits after injury increased significantly except at the first day; ④ HP variation in wounded fetal rabbits took a decrease tendency while that in wounded adults took an increase tendency. CONCLUSION: Adult rabbit skin is of higher capability of collagen synthesis than fetal rabbit skin. After wound, collagen synthesis in both fetal and adult rabbit skin increases. But the elevation in the former is temporary, suggesting rapid healing, and that in the latter is continuous, indicating a delayed healing and scar formation.
