Synergy between Staphylococcus Aureus and Pseudomonas Arenginosa in a Rat Model of Complex Orthopaedic Wounds

摘  要:目的研究动物接种和伴随金黄色葡萄球菌(以下简称金葡菌)和绿脓杆菌感染的互相作用.方法采用二组Sprague-Dawley鼠作动物试验.一组用金葡菌或绿脓杆菌接种,另一组接受可变浓度的两种实验微生物的复合菌接种.在接种后14 d处死,将来自伤口的标本用来培养.计算相关的每个接种体的感染率,以及将两项接种进行比较.结果 1D50对金葡菌是2.8×104 CFU,以及对绿脓杆菌是4.8×105 CFU.103 CFU的金葡菌与低浓度(102,103,或104 CFU)的绿脓杆菌结合产生的感染率,比单独任何一种相同的浓度感染率高的多.结合103 CFU的金葡菌和103 CFU的绿脓杆菌产生75%的感染率,比相关的103 CFU的任何一种微生物CFU高的多.绿脓杆菌的浓度增高时(C105,106和107 CFU),这种倾向则倒转,以及感染率则减少至33%(P=0.04).低浓度的绿脓杆菌(0~105 CFU)结合106 CFU的金葡菌产生83%~100%的感染率.在高浓度的绿脓杆菌(106及107CFU)减少感染率至33%(P=0.005).结论当伤口中存在每种低浓度细菌时,在金葡菌和绿脓杆菌之间的协同作用证明,当绿脓杆菌浓度增加时,感染率预期下降.在这种骨科切口模型中,认为低浓度的绿脓杆菌增加金葡菌感染的能力.同时,以这个率值的金葡菌试验,减少绿脓杆菌的感染率.Objective To study an interaction in animals inoculated concomitantly with staphylococcus aureus and pseudomonas aeragine-sa during a study of the efficacy of surfactants for disinfection of orthopaedic wounds. This led us to investigate whether synergy could be demonstrated between staphylococcus aureus and pseudomonas aeruginosa in a rat model of complex othopaedic wounds. Methods A wire was implanted into the spinous process of a lumbar vertebra of sprague - dawley rats through a clorsal incision. Animals were divided into two groups: group one was inoculated with either staphylococcus aureus or pseudomonas aeruginosa, and group two receired a polymicrobial inoeulation with both test orgaisms in varying concentrations. After inoculation, the wounds were irrigated and closed. On postoperative day 14, all animals were killed and specimens from the wound were cultured. The infection rate associated with eath inoculum combination was calculated, and two groups were compared. Results The 1D50 was 2.8 × 104 CPU for staphylococcus aureus and 4.8× 10 CFU for pseudomonas aeruginosa. The combination of 10 CFU of staphylococcus aureus with low concentrationg (10 , 10 , or 104 CPU) of pseudomonas aeruginosa yielded infection rates that were higher than those found with either organism alone at the same concentrations. The combination of 103 CFU of staphylococcus aureus and 103 CFU of pseudomonas aeruginosa yielded a 75% infection rate, which was significantly higher (P = 0.04) than that associated with 103 CFU of eitter organism alone. As the pseudomonas aeruginosa concentration was increased (to105, 106 , and 107 CPU). This trend reversed, and the infection rate decreased to 33% (P = 0.004) . Low Concentrations of pstudomonas aeruginosa (to 105 CFU) Combined with 106 CFU of staphylococcus aureus yielded infection rates ranging from 83 % to 100%.
