摘  要:目的 :采用基因技术 ,旨在从分子水平探讨骨仙片的作用机理。方法 :取 1 2 0只昆明小鼠造成桡骨1 .5 mm缺损骨折 ,随机分为两组 :服药组及未服药组各 60只 ,在术后 2 w及 4w,分别进行骨痂总 RNA的分离提取 ,通过与 、 型胶原质粒探针 ( p Mco1 1 a-1及 p Mco1 1 2 al-1 )斑点杂交 ,比较 型胶原 m RNA表达的变化。结果 : 型胶原 m RNA表达在术后 2 w明显小于 4w, 型胶原 m RNA表达术后 2 w明显大于 4w,提示 型胶原 m RNA表达代表骨形成及塑形的特征标志 , 型胶原 m RNA表达代表软骨修复的特征标志。 型胶原 m RNA表达量 ,服药组 >未服药组 , 型胶原 m RNA表达量 ,服药组 <未服药组 ,提示服药组骨形成迅速 ,由软骨修复期提早进入骨修复及塑形期。结论Objective:Gene technology is applied in this epxeriment for seeking molecular mechanism of `Gu-Xian' tablets.Methods:A totol of 120 mice,which were produced by operation a 1.5mm defect fracture in bilateral radius diaphysis,were divided egually into 2 proups (treatment and control) at random,mice in treatment group were taken with `Gu-Xian' tablets.Animals in two groups were killed respectively after operation of 2w and 4w,a total of RNA was extracted from callus of fracture site and was hybridized with probes of pMCol1a-1 and pMCol2a-1 in dot blot.Results:Whether taking CH(Chinese herb) or not,the expression level of Type Ⅰ collagen mRAN in 2w postoperation was apparently lower than in 4w,but the expression level of Type Ⅱ collagen mRNA in 2w postoperation was apparently higher than that in 4w.The experiment suggested the expression of Type Ⅰ collagen mRNA had characteristic of ossification and remodeling,while Type Ⅱ collagen mRNA had characteristic of cartilagious reparation.As compared the treatment group with control group,the expression value of Type Ⅰ collagen mRNA in the former was higher than that in the latter,Type Ⅱ collgen mRNA was opposite.It was fully proved that osteogenesis in treatment group was rapid increase,bone repair process entered into remodeling phase from cartilagious reparation was in advance.Conclusion:The molecular mechonism of `Gu-Xian' tablets to enhance and increase bone repair was clear.
