摘 要:目的 探讨原花青素 (Procyanidolic,pc)对急性呼吸窘迫综合征 (Acuterespiratorydistesssyndrome ,ARDS)大鼠疾病模型的治疗作用。方法 大鼠用百草枯 (Paraquat,PQ)一次性灌胃 ,剂量 2 5 0mg/kg,治疗组分别于染毒 6、2 4、4 8h各给予PC治疗剂量 5 0 0mg/kg ,染毒组与治疗组分别于 2 4、4 8、72h ,取大鼠血浆和支气管肺泡液 (BALF) ,测试其血浆和BALF中超氧化物歧化酶 (SOD)、丙二醛 (MDA)谷胱甘肽过氧化物酶 (GSH_Px)的活力 ,各组取肺病理组织做病理组织观察研究。结果 治疗组与染毒组比较 ,血浆和BALF中GSH_PX 、SOD活力逐渐升高 ,差异有显著性P <0 0 5 ,MDA含量下降 ,差异有显著性P <0 0 5 ,染毒与对照组比较 ,血浆和BALF中MDA含量高于对照组 ,差异有显著性P <0 0 5 ,SOD和GSH_PX 活力下降 ,差异有显著性P <0 0 5 ,肺组织病理学观察 ,ARDS组肺泡壁充血 ,炎症细胞浸润 ,Ⅰ型与Ⅱ型细胞破坏 ,肺泡内有炎症渗出物 ,对照组肺泡壁完好 ,肺泡内没有炎症渗出物 ,没有炎症细胞浸润 ;给予PC治疗后 ,肺泡壁充血、出血减轻 ,炎症渗出物减少 ;结论 PC对ARDS模型大鼠有一定的治疗效果。Objective To investigate the treament effect by procyanidine(PC) on pathologic model of rat with the acute respiratory distress syndrome(ARDS).Methods Rats were perfused using paraquat(PQ,250 mg/kg). In therapy group the rats were given by PC (500 mg/kg) at 6 h, 24 h and 48 h. After 24 h, 48 h and 72 h, blood plasma and broncho alveolas lavage fluid (BALF) were collected from the rats. The activity of SOD, MDA and GSH_P-X were tested in the blood plasma and BALF and the pathological tissues were observed. Results In comparison with ARDS model group, the activity of SOD and GSH_P-X were enhanced (%P%<0.05) in the blood plasma and BALF in PC therapy group, but MDA content obviously reduced (%P%<0.05). Compared with control grup, ARDS model group had raised MDA content (%P%<0.05) and lower activity of SOD and GSH_P-X(%P%<0.05). The alveolus wall of ARDS tissue was congested and immersed by inflammatory cells in the pathological observation of lung tissue. types Ⅰ and Ⅱ of the cell were broken and inflammatory substance was present in alveoli. But the alveolus wall was intact and had no inflammatory substance and cell in the lung tissue of the control group. With PC therapy, the alveolus signs of ARDS tissue were lightened about the aspects of congestion, bleeding and inflammatory substance.Conclusion PC therapy showed a definite effect on the ARDS model of rat.
