摘 要:通过亚临界水提取丹参药材中的丹参酮ⅡA的研究 ,建立了中药材中的脂溶性成分的亚临界水提取条件。分别考察了温度、压力、提取时间、提取物颗粒度等因素对提取量的影响 ,并与有机溶剂提取法比较。结果表明 :当两者具有相同的提取效率时 ,亚临界水提取法的提取时间及提取溶剂的消耗大大减少 ,并避免了因使用有机溶剂而造成的污染。提取最佳条件为 :样品颗粒度为 0 .18~ 0 .15mm ,5MPa ,180℃保持 10min。并通过提取 HPLC联机分析实现了对提取物的实时监测。A study was carried out for extraction of Tanshinone II A from Danshen with subcritical water to establish the method of extraction of liposoluble substances from the Chinese traditional 'medicine' with subcritical water. The influence of various parameters on the extraction efficiency, including the pressure, temperature and extraction time, as well as the particle size of the plant material was investigated. Compared with conventional organic solvent extraction, the subcritical water catraction shows lower handling time and solvent consumption with no waste produced at the same extraction efficiency. However, this technique is unsuitable for heat - sensitive compounds. The optimal condition of extraction for Tanshinone II A from Danshen with subcritical water is: 0.15 similar to 0.18 min particle size, 5 MPa, 180degreesC extraction for about 10 min. Furthermore, the online detection for extraction is achieved. This technique seems very promising for the extraction of heat-stable liposoluble substance from traditional medicine.
