摘  要:目的 研究四氢喋呤 (tetrahydrobiopterin ,BH4)对于胆固醇喂养兔的血管内皮舒张功能的影响。方法  2 4只新西兰兔随机分为三组 :A组进食普通饲料 ,B、C组进食高脂饲料。C组在指标观测前 2h耳缘静脉注射BH410mg/kg ,5周后三组动物取胸主动脉进行离体血管条实验 ,分别测定血管条对乙酰胆碱 (Ach)、三磷酸腺苷 (ATP)、硝普钠 (SNG)的累积浓度舒张反应。结果 B组血管条对Ach、ATP的最大舒张反应能力较A组明显降低 (分别为Bmax =4 65± 1 63比Amax =13 87±2 62 ,Bmax =4 2 7± 1 41比Amax =12 0 3± 2 55,P <0 0 1) ;C组对Ach、ATP的最大舒张反应能力较B组明显改善 (分别为Cmax =9 0 6± 2 14比Bmax =4 65± 1 63 ,Cmax =10 87± 1 89比Bmax= 4 2 7± 1 41,P <0 0 1) ;而三组血管条对硝普钠的最大舒张功能无明显改变 (Amax =7 2 0± 2 3 0 ,Bmax =6 77± 1 86,Cmax =6 2 3± 2 13 ,P >0 0 5)。结论 高胆固醇血症时内皮依赖性舒张功能受损 ,四氢喋呤能改善高胆固醇血症时内皮依赖性舒张功能 。Objective To investigate the influence of tetrahydrobiopterin on endothelium-dependent vasodilation in rabbits with high cholesterol diet Methods The rabbits received normal diet(A group,n=8) or cholesterol diet(B group, n=8) or cholesterol diet plus tetrahydrobiopterin(C group, n=8) Five weeks l ater, thoracic aortas were removed and cut into ring segments about 3 mm long, then the cumulating dose response to the endothelium-independent vasodilator sodium nitroprusside and the endothelium-dependent vasodilator Ach and ATP were examined Results The maximal relaxation of arterioles induced by Ach and ATP were significantly reduced in cholesterol group compared with those in control group (Ach Bmax=4 65±1 63, Amax=13 87±2 62,P<0 01);ATP Bmax=4 27±1 41, Amax=12 03±2 55 P<0 01) The maximal relaxation of arterioles induced by Ach and ATP were significantly enhanced in tetrahydrobiopterin group However, no significant changes in maximal relaxation of arterioles induced by sodium nitroprusside among three group s (Amax=7 20±2 30, Bmax=6 77±1 86, Cmax=6 23±2 13, P>0 05) Conclusion Rabbits with hypercholesterolemia may cause dysfunction of endothelium-dependent vascular relaxation, tetrahydrobiopterin could restore the response to endothelium-dependent vasodilation
