摘  要:应用放射分析方法 ,研究了3 H -GABA在小白鼠外周组织肾脏切片中的摄取 ,结果表明 ,小白鼠全肾切片对3 H -GABA的摄取是时间和浓度依赖性的 ,且是可以饱和的 ,当培育时间为 15min浓度时3 H-GABA摄取达到最高 ,而3 H -GABA浓度为 2 0 3nmol·L-1时 ,3 H -GABA的摄取也达到最高 (P <0 0 5 ,n =6~ 7) ,另外 ,药物对全肾切片对3 H -GABA的摄取也有影响 ;青霉素钾、苯巴比妥钠、苦味毒、氯苯氨丁酸、蝇蕈醇均使3 H -GABA的摄取明显减少 (P <0 0 5 ) ,而荷包牡丹碱却明显地提高3 H -GA BA的摄取 (P <0 0 5 )。结果提示 :小白鼠外周组织中同样存在GABA受体及GABA摄取系统 。Method of radio-Ligand was used in this paper. An uptake 3?H-GABA occurred in the whole kidney segment(0.2 mm in thickness)of mouse, segment were incubated at 36℃ in a medium containing 3?H-GABA which was show to be concentration-dependent, time-dependent and may have been saturated. When segments were incubated for 15 min, accumulation of 3 ?H-GABA was higher than that at 5 min and 10 min. When segment were incubated in 3?H-GABA(20 3 nmol/L),uptake of 3 H-GABA was higher than that at 3.85nmol/L and 7 7nmol/L. Benzylpenicilline kalium (250 μmol/L),Phenobarbitalum Natrium (250 μmol/L), picrotoxin(250 μmol/L), Baclofen(250 μmol/L) and Muscimol(125 μmol/L) caused significant decrease in the accumulation of 3?H-GABA in whole kidney segment of mouse in contrast, Bicuculline(250 μmol/L)resulted in a marked increase in the uptake of 3?H-GABA in whole kidney segment of mouse.
