6种增溶剂对棉铃虫细胞色素P450 O-脱甲基活性的影响

摘　　要：比较了 6种增溶剂对棉铃虫细胞色素P4 5 0O 脱甲基活性的影响。结果表明 ,在匀浆缓冲液中加入Polyoxyethyleneetherw 1,棉铃虫细胞色素P4 5 0的O 脱甲基活性为 37 80U (mOD·min-1·larva-1) ,是对照的 1 34倍 ;经Chaps、胆酸钠、Emulgen 911、Emulgen 913、Triton X10 0增溶处理后 ,O 脱甲基活性分别为 2 2 7,2 7 5 8,9 6 4 ,14 77和 19 86U ,分别是对照的 97% ,98% ,34% ,5 2 %和 70 %。对照中只有 79 2 %的个体能够被检测到活性 ,而在Polyoxyethyleneetherw 1处理中 10 0 %的个体都能够被检测到活性。因此Polyoxyethyleneetherw 1可以作为棉铃虫抗药性生化机理及生化检测、细胞色素P4 5 0分离纯化等方面研究的有效增溶剂。The effect of six cosolvents on O-demethylase activity of cytochrome P450 to P-NA was assessed by microtier plate kinetic assay.The mean O-demethylase activity of cytochrome P450 by solubilization with Polyoxyethylene ether w-1 is 37.80 U(mOD·min -1·larva -1),which is about 1.34 times that of the control(without adding any cosolvent).The mean activities of O-demethylase activity of cytochrome P450 solubilized with the other cosolvents such as Chaps,Cholic acid,Emulgen 911,Emulgen 913,Triton-X100 are 22.7,27.58,9.64,14.77 and 19.86 U,which are 0.97,0.98,0.34,0.52 and 0.7 times that of the control respectively.The results suggest that Polyoxyethylene ether w-1 can be used as an effective cosolvent in purification and biochemical detection of cytochrome P450 in Helicoverpa armigera.
