摘 要:采用RAPD技术对15种猕猴桃种质资源进行鉴定.结果表明,材料选用1年生休眠枝条,取刚展叶1~2d的幼叶,用改良CTAB法提取DNA较好;在所选的引物中,S21对11个品种中的7个样品的RAPD指纹图谱鉴定效果较好,可以以此作为鉴定小果甜、华光2号、海沃德等品种的特征指纹图谱,并将其中的7个品种分成2组,一组为大扩增片段组,包括小果甜、华光2号、海沃德.第2组为小扩增片段组,包括华美2号、金魁、米良1号、徐香,海沃德被划分到了大片段组即中华猕猴桃组内.S130扩增带数较多,可作为至少6个以上猕猴桃品种资源鉴定中的特异性引物.S269,S218引物也都有特异的带谱,但特异性不很强.The RAPD technology was used to identify the germplasm resources in 15 varieties of actinidia.The Results showed that it was better to extract DNA with CTAB method from young leaves of 1~2 days of dormant branches. Among the chosen primers, the effect of primer S21 is better on the fingerprint pattern identification of 7 out of the 11 samples and could be used to identify the characteristic fingerprint patterns of Xiaoguotian, Huaguang No.2 and Hayward. And the 7 of them could be broken into 2 groups, one group of larger segments, including Xiaoguotian , Huaguang No.2 and Hayward, the other group of smaller segments including Huamei No.2,Jinkui ,Miliang No.1,Xuxiang ,among which Hayward was classified into the group of larger segments ,i.e. the Chinese group .And primer S130 produced more amplification bands ,and could be used as the special primer in the identification of at least 6 Kiwifruit gremplasm resources. Primer S269 and S218 had their own special band patters, but their speciality was not very strong.
