摘  要:目的研究严重创伤早期大鼠下丘脑热休克蛋白70(HSP70)与诱导型一氧化氮合酶(iNOS)转录表达变化分布和意义。方法采用BIM-Ⅲ型生物撞击机致大鼠胸部严重撞击伤,并造成单侧股骨骨折,运用S-ABC免疫组化、原位杂交技术测定下丘脑HSP70与iNOS及其mRNA的转录表达水平。结果正常对照组HSP70低水平表达,在伤后1h即开始明显增高(P<0.05),6h达到峰值为正常的18倍,12h后开始下降,24h时主要集中在室旁核内;iNOS无基础表达,在伤后1h开始出现,随HSP70同步增高,8h达到高峰,较1h时增加13倍。两者相关系数r=0.97601。结论严重创伤后早期下丘脑内HSP70和iNOS过度表达,在严重创伤应激时下丘脑的损伤与抗损伤机制中起重要作用。Objective To study the distinct distribution and the time-course changes of HSP70and inducible NOS expressions in the hypothalamua of rats following severe trauma.Methods The severe traumatic model that severe thorax impact injury with fracture of femur was established in rat.The ex pressions of HSP70and iNOS were studied with S-ABC histochemistry,and their mRNAs were detected with in situ hybridization.Results The expression of HSP70in control group was at low basic level.In the trauma group,it was increasing quickly from1h(P<0.05)after injury and came to the peak at6hours(increased17-fold),the positive cells mainly distributed in the paraventricular nucleus of hypothalamus(PVN)at24hours.The positive cells of iNOS couldn't be discovered until one hour after injury,then the number was increased alone with HSP70and reached the peak at8hours(increased13-fold).The coefficient of product-moment correlation was0.97601.Conclusion Severe trauma stress can cause the overexpressions of HSP70and iNOS in hypothalamus,which play an important role in the balance of injury and anti-injury in the hypothalamus stress reaction.[
