细胞凋亡对尖锐湿疣患者外周血单个核细胞的影响

摘　　要：目的 :探讨激活诱导细胞死亡 (AICD)对尖锐湿疣患者细胞免疫功能的影响。方法 :分离CA患者和健康体检正常人的外周血单个核细胞 (PBMC) ,在PHA的刺激下体外培养 2 4小时后收集细胞和上清液 ,经流式细胞术检测凋亡细胞 ,采用酶联免疫吸附试验检测培养上清液IL 2及IL 10。结果 :发现CA组新鲜血PBMC的凋亡率很低和对照组相似 (P >0 0 5 ) ,培养 2 4小时后CA患者的外周血单个核细胞凋亡明显高于正常对照组 (P <0 0 0 1) ,CA患者的培养上清液IL 2浓度明显低于对照组 (P <0 0 0 1) ,而IL 10浓度显著高于对照组 (P<0 0 0 1)。结论 :AICD可能是引起尖锐湿疣患者细胞免疫功能低下的一个重要机制 ,在尖锐湿疣患者发病机制中起重要作用。Objective:To investigate activation-induced cell death(AICD) to effect of cellular immune function in the condyloma acuminatum(CA).Methods:Peripheral blood mononuclear cells(PBMC) were isolated from normal healthy physical examination's persons(normal control group) and patients with CA.PBMC were cultured with PHA-P for 24 h in vitro.Apoptosis of the PBMC was detected by flow cytometry.Supernatant's cytokines(IL-2 and IL-10) were assayed by ELISA.Results:The rate of apoptosis both CA group and control group in fresh PBMC was very low and similar(P>0.05).The rate of PBMC apoptosis with CA group was noticeably increased compared to that of normal control group(P<0.001) after PBMC were cultured for 24 h.The level of IL-2 was obviously lower in CA group than that in control group (P<0.001).The level of IL-10 was significantly higher in CA group compared to control group in culture supernatant(P<0.001).Conclusion:This study indicated that AICD may be an important mechanism of CA patient low cellular mediated immune function,which may play an important role in the pathogenesis of CA.
