摘  要:目的 以天然型无机骨 (NNB)为支架材料 ,复合体外诱导培养的骨髓间充质干细胞 (MSCs) ,构建组织工程化骨 ,并进行动物自体异位成骨实验 ,以期为骨缺损的修复提供较为理想的方法。方法 贴壁法培养兔MSCs ,体外诱导培养扩增。以 5 0× 10 6/ml的密度与NNB复合 ,构建组织工程化骨 ;部分材料体外培养 1天 ,电镜观察细胞与材料复合情况。其余以NNB为空白对照 ,分别植入 6只新西兰兔皮下 ,每组 6例 ,4周取材 ,脱钙组织学观察。结果 复合后 1天 ,细胞可在NNB内表面良好贴壁 ;植入动物自体皮下 4周 ,实验组材料网孔中有新生骨、软骨样组织形成 ;而NNB空白对照组材料内无新骨形成。结论 NNB具有良好的生物相容性 ,可为种子细胞生长提供理想的三维空间 。Objective To evaluate the tissue engineered bone constructed by in vitro cultured marrow cells combined with natural nonorganic bone(NNB) scaffold in rabbit subcutaneously.Methods MSCs were cultured and induced in vitro and combined with NNB with a concentration of 50×10 6/ml to construct tissueengineered bone. Some of the engineered bone were cultured in vitro for 1 day and tested the biocompatibility by scanning electron microscope(SEM). Others were implanted into 6 rabbits subcutaneously, adopting NNB as the control group(6 cases each group). Specimens were harvested 4 weeks after operation. Results MSCs would attached to the inner surface of NNB in 1 day after combination. It was showed in vivo study 4 weeks after operation, the new cartilage and new bone formed inside the scaffold in experimental group, which were not found in NNB control group.Conclusion The NNB had good biocompatibility, and provided ideal three dimension space for MSCs. It could be successfully used to construct tissueengineered bone.
