摘　　要：目的 :探讨Ito细胞在大鼠肝再生中的作用。方法 :分离大鼠 2 / 3肝切除术后不同时间 ( 6h、12h、2 4h)以及未切除肝的Ito细胞 ,提取其总RNA ,采用半定量RT -PCR方法检测其肝细胞生长因子 (HGF)mRNA的表达。结果 :分离的Ito细胞纯度达 91%。肝 2 / 3切除术后大鼠 6h、12h和 2 4h分离的Ito细胞HGFmRNA的表达较未切除组分别升高了 14 .6倍、2 1.1倍和 11倍。结论 :Ito细胞在肝大部分切除大鼠早期表达HGFmRNA升高 ,表明其在肝再生启动过程中具有重要作用。Objective To explore the role of Ito cells in liver regeneration of rats. Methods Ito cells were isolated from the rats at different times(6h、12h、24h)after 2/3 partial hepatectomy as well as the ones without hepatectomy.Total RNA was extracted from isolated cells.Then,hepatocyte growth factor(HGF) mRNA levels were semiquantitatively assessed by a RT-PCR method. Results The purity of islated Ito cells was 91%.We found that the level of HGF mRNA in Ito cells from rat liver rose 14.6 fold,21.1 fold and 11 fold within 6h,12h and 24h after hepatectomy repectively. Conclusion:Expression of HGF mRNA increases in hepatic Ito cells in early stage after 2/3 partial hepatectomy,suggesting that Ito cells play an important role at the initiation of liver regeneration.
