摘  要:目的 分析职业性慢性轻度苯中毒工人外周血单个核细胞中信号结合T细胞受体删除DNA环 (sjTRECs)的含量和CD4+/CD8+比值 ,从而了解其初始 (naive)T细胞含量和胸腺近期输功能 ,以及外周T细胞亚群分布情况。方法 利用间接免疫荧光法检测 11例健康人和 5例轻度苯中毒患者外周血单个核细胞中CD4+/CD8+比值 ;采用实时荧光定量聚合酶链反应 (real timePCR)和TaqMan方法检测两组外周血单个核细胞中sjTRECs的含量。结果 每 10 0 0个外周血单个核细胞中sjTRECs含量在正常人中为 9 10± 4 5 5 ,轻度苯中毒为 2 80± 3 79,P <0 0 5 ;轻度苯中毒者CD4+/CD8+的比值 (0 92± 0 3 5 )明显低于正常人 (1 49± 0 2 1) ,P <0 0 1。结论 轻度苯中毒患者胸腺近期输出功能低下 。Objective To analyze the content of signal joint T cell receptor excision DNA circles ( sjTRECs ) and the ratio of CD4 +/CD8 + within peripheral blood mononuclear cells (PBMCs), thereby to evaluate the content of naive T cells, the recent thymic output function, and the distribution of T cell subset in chronic mild benzene poisoning in comparison with normal individuals. Methods Quantitative detection of sjTRECs in DNA of PBMCs from 11 normal individuals and 5 patients with mild poisoning of benzene were preformed by real time PCR using TaqMan technique. The ratio of CD4 + and CD8 + were assessed by indirect fluoroimmunoassay. Results The mean value of sjTRECs was 9 10± 4 55/1?000 PBMCs in normal individuals and 2 80± 3 79/1?000 PBMCs in the patients respectively( P <0 05), and the mean value of CD4 +/CD8 + in patients was significantly lower(0 92±0 35) than that of controls(1 49±0 21)( P <0 01). Conclusion The results indicated that the recent thymic output function might decrease in patients with chronic mild benzene poisoning. Low dose exposure to benzene might cause cellular immunity disorder.
