摘 要:目的:探讨CTLA4Ig基因能否在移植小肠局部转染表达。方法:移植前经肠系膜上动脉给供肠灌注脂质体包裹的CT-LA4IgcDNA重组质粒,应用免疫组织学及逆转录多聚酶链反应(RT-PCR)观察移植小肠中CTLA4Ig转基因产物的表达。结果:免疫组织学及RT-PCR结果显示在移植后48h移植小肠中有大量CTLA4Ig转基因产物的表达。结论:经肠系膜上动脉体外灌注脂质体包裹的CTLA4IgcDNA重组质粒可有效转染移植小肠。Objective:To evaluate the possibility of local expression of C TLA4 Ig gene in small bowel allografts. Methods::The donor small bowels were perfus ed ex vivo with the reconstructed plasmid of CTLA4Ig cDNA packaged with lipofect in vector via intra-superior mesenteric artery before transplantation. The CTLA 4Ig transgene expression in the small bowel allografts was assessed by immunohis tology and reverse transcriptase polymerase chain reaction (RT-PCR) after trans plantation. Results:Immunohistology and RT-PCR demonstrated abundant expressio n of CTLA4Ig transgene in the allografts 48 h after transplantation. Conclusion :A single ex vivo intra-superior mesenteric artery infusion of reconstructed p lasmid of CTLA4Ig cDNA packaged with lipofectin can induce efficient transductio n of the small bowel allografts.
