摘　　要：目的　研究pcDAN3.1·PTH治疗甲状旁腺功能低下症时血钙的变化及血钙与 pcDAN3.1·PTH剂量的关系。方法　手术去掉家兔甲状旁腺 ,建立甲状旁腺功能低下症模型。将已构建的人甲状旁腺激素基因 pcDAN3.1·PTH以 10 0 ,30 0 ,5 0 0 μg/ml剂量注射模型家兔的骨骼肌内 ,测定注射后不同时间的血钙。 结果　注射pcDAN3.1·PTH后 2 4h时模型家兔的血钙升高 ,7d达高峰水平 ,并持续 2个月 ,以 30 0 μg/ml和 5 0 0μg/ml为佳。 结论　人甲状腺激素基因治疗甲状旁腺功能低下是有效的 ,且以 30 0 μg/ml和 5 0 0 μg/ml为最佳剂量。Objective:To measure the concentration of serum calcium in rabbits with hypoparathyrodism (HPT) which were injected with pcDNA3.1·PTH and study the relationship between the changes of serum calcium and pcDNA3.1·PTH dosage.Methods:Different dosages of pcDNA3.1·PTH(100?μg/ml,300?μg/ml and 500?μg/ml)containing human PTH gene were injected respectively into muscle of the experimental rabbits after parathyroidectomy in advance. Then the concentrations of PTH in blood of rabbits were measured at following different monents.Results:Serum calcium level began to increase from the first day, which reached peak concentration at the seventh day after the injection of pcDNA3.1·PTH and persisted for two months.Conclusions:Human PTH gene was effective in treating HPT rabbits and their best effective dosages were 300?μg/ml and 500?μg/ml.
