反相高效液相色谱法测定血清中卡铂浓度

摘　　要：建立了人血清中卡铂的反相高效液相色谱测定法。固定相为Shim—packCLC—ODS柱,重蒸馏水作流动相,检测波长为229nm。卡铂平均绝对回收率为99.61％,日内和日间平均变异系数分别为4.18％和5.23％。最低检出浓度为0.05μg/ml,线性范围0.1～100μg/ml(r=0.9999)。应用本法测定了4例肿瘤患者静滴卡铂后的血药浓度。A simple, rapid, accurate and precise reversed-phase HPLC method has been developed for the determination of carboplatin in human serum using Shim-pack CLC-ODS column (150mm×6mm ID). The mobile phase was distilled water and the eluent was monitored at 229 nm. Carboplatin elated at 6.1 min. The mean absolute recovery was 99.61％. Within day and clay-to-day precision (cv)were 4.18% and 5.23％, respectively. The measurable lowest level was 0.05ug/ml and the standard curve was linear over the concentration range of 0.1 100ug/ml (r=0.9999). The pharmacokinetic study of carboplatin in four cancerous patients was presented.
