摘  要:目的:观察烧伤小鼠早期肝组织JNK1及其磷酸化的时相变化特点,初步探讨烧伤后应激的分子机制。方法:将36只BALB/C小鼠随机分为正常对照组和实验组,实验组给予背部Ⅲ°15%-20%烧伤。分别于伤后2h、4h、6h、12h、24h用western blot测定肝组织JNK1的蛋白表达及其磷酸化水平。结果:烧伤后各时相点肝组织JNK1在蛋白水平与正常对照相比没有统计学意义的变化;而烧伤后早期(2h)JNK1磷酸化程度显著增加,4h达到高峰,6h时仍显著高于正常对照组水平(P<0.01),以后逐渐恢复至正常。结论:烧伤早期引起小鼠肝组织JNK1的磷酸化程度显著增强,而蛋白表达无明显变化。Objective:To observe the dynamic change of hepatic c-Jun NH2-terminal kinase 1(JNK1) and its phosphorylation status during the early stage in burned mice. Methods: Thirty-six BALB/c mice were randomized into control and trial group. After clipping of the hair, 15% - 20% thickness third degree burn injury was produced on the back of trialed mice. At 2,4,6, 12,24h after bum, the protein expression of hepatic JNK1 and its phosphorylation were detected by western blot. Results: There was not significant difference in the protein level of hepatic JNK1 at every stage between the control and trial group.The phosphorylation status of JNK1 was increased distinctly at the very early stage(2h) .reached the peak at 4h, and was still significantly higher than those in the normal control at 6h ( P < 0.01) ,then return to the normal level gradually.Conclusion:The hepatic JNK1 phosphorylation is significantly up-regulated after burn injury, yet there is not significant change in the protein expression.
