摘 要:目的观察高浓度葡萄糖对体外培养人晶状体上皮细胞SRA01/04增殖、周期和迁移的影响,筛选最佳增殖模型条件,为后续研究糖尿病性白内障的发病以及治疗机制提供必要的实验基础。方法将人晶状体上皮细胞SRA01/04分为正常对照组(0mmol·L-1)和不同高浓度葡萄糖组(浓度分别为5、10、15、2、25、30、35、40、45、50、55、60、65、75、100mmol·L-1),采用CCK-8法检测细胞的增殖,流式细胞术检测细胞周期,划痕实验评价细胞迁移能力。结果高糖作用下晶状体上皮细胞的增殖活力高于正常对照组,随着葡萄糖浓度的增加,细胞增殖活性在30mmol·L-1出现峰值,随着高糖暴露时间的延长,在48h时细胞的增殖活力更加显著,并能使G0/G1期细胞比例减少(P<0.05),S期比例增加(P<0.05);高糖组迁移的细胞数较正常对照组显著增强(P<0.05)。结论高浓度葡萄糖可影响人晶状体上皮细胞周期,促进细胞增殖,加强细胞迁移;以30mmol/L葡萄糖作用48h效果最显著,可作为制备细胞模型的条件。Objective To investigate effect of the different concentrations of glucose medium on expression of proliferation,cycle and migration in cultured Human lens epithelial cells and to provide necessary experimental basis for the follow- up study of patnogenesis and treatment mechanism of diabetic cataract. Methods Human len epithelial cell lines SRA01 /04 were cultured in vitro. The cells were exposed to either 0. 0( normal control group),or 5. 0,10. 0,15. 0,20. 0,25. 0,30. 0,35. 0,40. 0,45. 0,50. 0,55. 0,60. 0,65. 0,75. 0,and 100. 0mmol·L- 1glucose for 12,24,and 48 h. The survival and proliferation of the cells were quantified by CCK8 assay. Cell cycle analysis was performed by flow cytometry. Scratching wound assay evaluate the ability of cell migration. Results Compared with normol control group,high- glucose induced obvious proliferation of the SRA01 /04. With the extension time of high- glucose exposure,it showed notable proliferation in 48 h. Especially,the peaks of cell proliferation were in the group of 30. 0mmol / L glucose at 48 h.High- glucose( 30mmol·L- 1) induced decrease of cells in G0/ G1phase( P < 0. 05),but increased cells in S phase( P < 0. 05). Deducting cell proliferation by the cell viability ratio,the number of migrating cells in high- glucose group was significant increased compared with normal control group( P < 0. 05). Conclusion The different concentrations of glucose may increase the cell proliferation especially 30. 0mmol·L- 1glucose at 48 h. High glucose may promote the migration and assisted in the cycle at G1/ S phase of human lens epithelial cell. It is appreciate to choose 30. 0mmol·L- 1glucose at 48 h as the best concentration for establishing model.
