摘  要:目的探讨灯盏花素注射液对大鼠出血性脑损伤神经细胞凋亡的保护作用。方法用Ⅶ胶原酶联合肝素注入大鼠尾壳核建立脑出血模型,随机将大鼠分为空白对照组、假手术组、模型组和治疗组,治疗组于术后注射灯盏花素注射液(2mg·kg-1)后第3、5、7天后取材,应用Berdson评分标准对各组大鼠进行神经功能评分,应用Neu N/TUNEL荧光法检测出血灶周围神经元凋亡,应用流式细胞术检测神经细胞凋亡率,应用免疫组织化学方法检测出血灶周围细胞Bcl-2、Bax蛋白的表达。结果脑出血模型组及灯盏花素治疗组各时间点均出现偏瘫症状,与脑出血模型组各相同时间点比较,灯盏花素治疗组大鼠神经功能评分明显减低(P<0.05)。Neu N/TUNEL荧光染色:脑出血组及灯盏花素治疗组出血灶周围细胞核仁固缩、边集,与脑出血模型组比较,灯盏花素治疗组各时间点脑出血灶周围Neu N/TUNEL阳性细胞明显减少(P<0.01)。流式细胞术检测结果:与脑出血模型组比较,灯盏花素治疗组各时间点脑出血灶周围神经细胞凋亡率明显减少(P<0.05);免疫组化结果显示:与脑出血模型组相比,灯盏花素治疗组各时间点出血灶周围Bcl-2阳性细胞数量明显增多(P<0.05),而Bax阳性细胞数量明显降低(P<0.05)。结论灯盏花素注射液对大鼠出血性脑损伤的保护作用可能通过促进Bcl-2蛋白,抑制Bax蛋白的表达,抑制出血灶周围神经细胞的凋亡,促进神经功能恢复。Objective To explore the protective effect of breviscapine injection on apoptosis of intracerebral hemorrhage in rats. Methods 80 healthy SD rats were randomly divided into blank control group,sham operative group,cerebral hemorrhage model group and breviscapus treatment group. Cerebral hemorrhage model was established in rats with a mixture of collagenase and heparin injected into caudate-putamen. Rats in cerebral hemorrhage model group and breviscapus treatment group were killed on the 3rd,5th and 7th day after drug injection. The neurological functional scores of each group were evaluated according to the Berdson Scoring Criteria. Apoptotic cells were detected by TUNEL fluorescence method and apoptotic rate of nerve cells was determined by flow cytometry. The expression of bcl-2 and bax around hemorrhagic focus was tested by immunohistochemical methods. Results Hemiplegic symptom appeared in cerebral hemorrhage model group and breviscapus treatment group at each time point. Compared with cerebral hemorrhage model group at the same time point,neurological score of breviscapus treated rats was significantly lower( P < 0. 05). In TUNEL florescence staining,perihematomal cells exhibited nucleolus pyknosis and margination,and compared with cerebralhemorrhagemodelgroup,perihematomalTUNELpositivecellsdecreased significantly in the breviscapine treatment group at each time point( P < 0. 01). The apoptotic rate of perihematomal brain cells were tested by flow cytometry analysis and showed that: compared with cerebral hemorrhage model group,apoptotic rate of cerebral perihematomal in breviscapine treatment group at each time point reduced significantly( P < 0. 05). Immunohistochemical detection showed that compared with cerebral hemorrhage model group,bcl-2-positive cells in breviscapine treatment group at each time point increased significantly( P < 0. 05),while bax-positive cells were significantly decreased( P < 0. 05). Conclusion Breviscapine can reduce the number of perihematomal TUNEL positive cells and apoptosis
