摘 要:目的探索一种新的瓣膜成形方法。方法用酶消化法获取人脐静脉内皮细胞并进行传代培养,绘制生长曲线;并对内皮细胞行Ⅷ因子抗原鉴定。用激光行心脏瓣膜成形,然后将内皮细胞种植在成形的瓣膜表面。共分4组,每组两块瓣膜,分别于培养后第2、4、6、8天取出,用扫描电镜观察瓣膜表面细胞生长情况;将培养8d的瓣膜放入体外循环装置中,经受培养液冲击4h后,用扫描电镜观察瓣膜表面细胞粘附情况。结果种植2d后,瓣膜表面约20%~30%被内皮细胞覆盖,4d后可达50%~60%,6d后可达70%~80%,8d基本形成内皮细胞单层。模拟正常循环血液冲击后,仍可见内皮细胞粘附在瓣膜表面。结论激光心脏瓣膜成形加内皮细胞种植是一种值得进一步临床应用探讨的方法。Objective To establish a new way of cardiac valvoplasty. Methods The endothelial cells from human umbilical vein(HUVEC) were obtained with enzymes,and passaged,finally to draw up cell growth curves and evaluate factor Ⅷ antigen. The valvoplasty was conducted with laser and the endothelial cells were seeded on the surface of formed valves.The study divided into 4 groups with 2 valves in each group.The valves were observed under scanning electron microscope after 2,4,6,8 days, respectively and culturing.The valves after 8 days culturing were washed by culture medium for 4 hours and observed under scanning electron microscope. Results After 2,4 and 6 days culturing, the valves were covered by HUVEC in 20-30%,50-60% and 70-80%,respectively. After 8 days,culturing the HUVEC formed a monolayer on the surface of the valves near totally.There were still a lot of HUVEC on the valve after washing. Conclusions Lasers valvoplasty and endothelialization of the valves are proved to be worthy for further clinical trial.
