摘  要:[目的]研究一定浓度镉离子诱导离体SD大鼠肾细胞的凋亡及引起细胞内钙稳态的失调 ,并分析亚硒酸钠对镉离子上述作用的影响。[方法]在37℃、5 %CO 2 条件下 ,离体SD大鼠肾细胞分别在含不同浓度镉离子 (150、75、30、15、0μmol/LCdCl2)和硒加镉离子 (1.15μmol/LNa2SeO3+30μmol/LCdCl2)的FD培养液中孵育3h后 ,用吖啶橙/溴乙锭双荧光染色法检测细胞的凋亡率 ,以fluo_3/AM为探针研究细胞内游离钙离子浓度的变化。[结果]不同浓度的镉离子能不同程度地诱导SD大鼠肾细胞的凋亡 (15~150μmol/LCdCl2 各组凋亡率分别为57.5 %、45.9 %、39.3 %、34.7 % ) ,同时还能引起不同程度的细胞内游离钙离子浓度增加 ,并呈现剂量效应正相关。低浓度的亚硒酸钠有一定的降低镉离子诱导细胞凋亡的作用 (硒加镉组的凋亡率仅为19.5 % )。Objective To study apoptosis and disequilibrium of calcium homeostasis induced by cadmium in SD rat kidney cells,and to analyse this processes being influenced by the sodium selenite.[Methods] Under the condition of37℃and5%CO 2 ,SD rat kidney cells were incubated in FD medium at different concentrations of cadmium(150,75,30,15,0μmol CdCl 2 /L)and cad-mium with selenite(1.15μmol Na 2 SeO 3 /L+30μmol CdCl 2 /L)for3h,then the apoptosis was detected by using acridine orange/ethidiumbromide double fluorescent dye staining,and the free calcium concentration in cells was detected by using fluorescent indicator fluo-3/AMas the probe.[Results] The different degree apoptosis in SD rat kidney cells was induced at different concentrations of calcium,the rate of apoptosis in groups150~15μmol CdCl 2 /L was57.5%,45.9%,39.3%,34.7%,respectively.The result showed that the elevation of the comcentration of cadmium caused an increase in rate of apoptosis and in calcium concentration in cells in a dose-dependent positive correlation.The results also showed that sodium selenite in lowconcentration can decrease the rate of apoptosis induced by cadmium,the rate of apoptosis in the groups of cadmium with selenite was19.5%.[Conclusion] Cadmium might cause apoptosis and disequilibrium of calcium homeostasis in SD rat kidney cells.
