蠲哮片对哮喘豚鼠sICAM-1蛋白水平及ICAM-1基因表达的影响

摘　　要：目的 :探讨蠲哮片治疗支气管哮喘的机理。方法 :参照Huston方法建立豚鼠哮喘模型 ,采用ELISA方法检测正常组 ,模型组 ,蠲哮片高、中、低各剂量组的sICAM 1蛋白水平 ;应用RT PCR技术观察蠲哮片对各组豚鼠肺组织ICAM 1基因表达的影响。结果 :( 1 )模型组与各组之间血浆sICAM 1含量比较 ,有极显著性差异 (P <0 .0 1 ) ,且随着蠲哮片的剂量的增加 ,血浆sICAM 1含量明显降低 ;( 2 )正常对照组肺组织ICAM 1mRNA表达量极低 ,模型组ICAM 1mRNA表达量明显增高 ,应用蠲哮片后其表达逐渐降低 ,且随着剂量增加降低程度增加。结论 :蠲哮片可降低哮喘豚鼠血浆sICAM 1蛋白合成 ,可降低哮喘豚鼠肺组织ICAM 1mRNA表达量 ,其治疗哮喘的机理可能是通过降低ICAM 1的逆转录水平 ,进而减少ICAM 1蛋白合成 。Objective:To study the mechanism of Juanxiao Tablet(JXT ) in treating asthma. Methods:To set up the guinea pig asthmatic model consultin g Huston's method and assay soluble intercellular adhesion molecule 1(sICAM 1) level in plasma by enzyme linked immunosorbent assay(ELISA),investigate JXT's i nfluence on ICAM 1mRNA gene expression by reverse transcription polymerase cha in reaction(RT PCR) technique.Results:Compared with the model group,there were three kinds of changes in the larger quantity of JXT treated group. ① The level s of sICAM 1 in phasma were decreased significantly (from 47.68±18.41,61.83±16 .17,74.37±21.06 to 153.14±76.29,respectively, P <0.01). ②The ICAM 1 expre s sion in lung tissues was inhibited.Conclusion:JXT can decrease the guinea pigs' airway byperresponsiveness(AHR) which may associate with that JXT plays a role i n inhibiting the expression of ICAM 1 and lowering ICAM 1's synthesis\;
