A Method to Isolate and Culture Highly Purified Populations of Glandular Epithelial and Stromal Cells from Human Endometrial Specimens

摘  要:【目的】探索一种简便、高纯度分离在位子宫内膜腺上皮细胞和间质细胞的方法,建立高纯度体外子宫内膜细胞培养模型。【方法】选择正常的新鲜子宫内膜组织20例,通过酶解,过滤及贴壁纯化等技术,每例均用Ryan方法(对照组)及改良的新方法(改良组)分离、纯化及培养子宫内膜腺上皮细胞及间质细胞,并进行传代。【结果】18例标本获得成功,对照组分离的间质细胞纯度为(84.22±5.17)%,腺上皮细胞纯度为(62.11±6.23)%;改良组分离的间质细胞纯度达(97.89±0.96)%,腺上皮细胞纯度(95.12±2.11)%,与对照组比较,差异有显著性意义(P<0.01)。【结论】改良方法简便,可高纯度分离子宫内膜腺上皮细胞及间质细胞。在体外建立高纯度子宫内膜细胞培养模型,更利于在细胞和分子水平上研究子宫内膜的各种功能及干预调节。To investigate an improved method for simple and highly purified isolation of endometrial glandular epithelial cells and stromal cells for in vitro culture 20 fresh endometrial specimens were collected from the operating theater and transferred to the tissue culture laboratory which were histologically normal. In investigated group and control group, Enzymolysis, filtration and pasted wall purification were used to isolate, purify and culture the endometrial stromal and glandular cells and reproduce.The endometrial cell cultures were successfully established in 18 of 20 endometrium samples . The stromal cells purity was(97.89±0.96)%and glandular epithelial cells (95.12±2.11)%by the improved method in investigated group,while the purities of control were(84.22±5.17)%and(62.11±6.23)%respectively (P< 0.01). [Conclusion]An improved method is successfully developed for simple and highly purified isolation of endometrial glandular epithelial cells and stromal cells for in vitro culture. Culture of these cell populations will help study their functions and molecular regulations.
