摘 要:目的 :探讨心痛灵滴丸预适应对兔缺血再灌注损伤心肌细胞凋亡及相关基因Bcl- 2、Bax蛋白表达的影响。方法 :建立冠状动脉粥样硬化 (AS)家兔在体心肌缺血再灌注模型 ,采用原位末端TUNEL法检测心肌细胞凋亡指数 (AI) ,免疫组化S -P法检测Bcl- 2、Bax蛋白表达。结果 :缺血再灌注时AI增加 ,Bcl- 2蛋白表达减弱 ,Bax蛋白表达增强 ,心痛灵滴丸预适应和缺血预适应都能使AI和BaxOD值明显下降(P <0 0 1) ,Bcl- 2OD值上升 (P <0 0 1)。而且心痛灵滴丸预适应能加强缺血预适应对兔缺血再灌注心肌细胞凋亡的抑制作用。结论 :心痛灵滴丸预适应能抑制缺血再灌注损伤时心肌细胞凋亡 ,其机制可能与上调Bcl-Objective:To explore the effects of xintongling pill (XTL) preconditioning on apoptosis and Bcl-2,Bax expression in rabbits'myocardial cells during myocardial ischemic reperfusion(I/R). Methods:Myocardial ischemia reperfusion injury models were established in atherosclerosis (AS) rabbit by ligating the left descending anterior coronary artery for 30 min, followed by 120 min of reperfusion in vitro.Apoptosis was identified by TUNEL and apoptosis index (AI) was obtained.The protein expression of Bcl-2 and Bax genes was investigated by S-P immunohistochemical stain. Results:The AI was significantly increased during myocardial ischemic reperfusion,whereas protein expression of Bcl-2 gene was decreased and protein expression of Bax gene was increased.The AI and the average OD value of Bax protein significantly lowed,while the average OD value of Bcl-2 protein was significantly higher in XTL group and IPC group than that in I/R group (P<0 01).There was synergic effect on apotosis in AS rabbit myocardial cells between XTL preconditioning and ischemic preconditioning. Conclusion:XTL preconditioning can inhibit myocardial cells'apoptosis induced by ischemia reperfusion in AS rabbit.The mechanism may be associated with upregulating protein expression of Bcl-2 gene and downregulating protein expression of Bax gene.
