摘 要:目的探讨高氧损伤状态下,肺组织内表面活性物质相关蛋白A(SPA)、表面活性物质相关蛋白B(SPB)、表面活性物质相关蛋白C(SPC)mRNA的变化规律及其在肺水肿中的作用。方法将新生大鼠120只依据吸氧浓度(Fi O2)不同分为研究组1(Fi O280%)、研究组2(Fi O260%)、研究组3(Fi O240%)和空气对照组(Fi O221%),每组40只。各组分别于实验后1、3、5、7、14 d行反转录聚合酶链反应(RT-PCR)检测SPA、SPB、SPC mRNA,并对各组检测结果进行比较。结果各组新生大鼠肺组织内SPA mRNA水平在实验后1、3 d时比较差异均无统计学意义(P>0.05),研究组1和研究组2新生大鼠肺组织内SPA mRNA水平在实验后5、7 d时均较空气对照组低(P<0.05),研究组1在实验后5、7 d时和研究组2在实验后7 d时新生大鼠肺组织内SPA mRNA水平均较研究组3低(P<0.05),研究组1新生大鼠肺组织内SPA mRNA水平在实验后14 d时高表达,较空气对照组显著升高(P<0.05)。各组新生大鼠肺组织内SPB mRNA水平在实验后1、3 d时比较差异均无统计学意义(P>0.05),研究组1和研究组2在实验后5 d时新生大鼠肺组织内SPB mRNA水平显著低于空气对照组,研究组1在实验后7、14 d时均较空气对照组显著升高,且14 d时较研究组3也明显升高(P<0.05)。各组新生大鼠肺组织内SPC mRNA水平在实验后1、5 d时比较差异均无统计学意义(P>0.05),研究组1、研究组3新生大鼠肺组织内SPC mRNA水平在实验后3 d时较空气对照组显著降低(P>0.05),研究组1在实验后7、14 d和研究组2在实验后14 d时新生大鼠肺组织内SPC mRNA水平虽有升高趋势,但仍显著低于空气对照组(P<0.05),研究组1新生大鼠肺组织内SPC mRNA水平在实验后7 d时较研究组2低、14 d时较研究组3低(P<0.05)。结论 SPA、SPB、SPC mRNA在高氧肺损伤早期表达明显降低,可能是导致肺水肿的重要原因之一。Objective To study the change regularity of surfactant associated protein A( SPA),surfactantassociated protein B( SPB) and surfactantassociated protein C( SPC) mRNA in the lung tissues with injury induced by hyperoxia and explore their role in pulmonary edema. Methods According to the fraction of inspired oxygen( Fi O2),one hundred and twenty newborn rats were divided into group 1( Fi O280%),group 2( Fi O260%),group 3( Fi O240%) and air control group( Fi O221%),with 40 rats in each group. The expression of SPA,SPB,SPC mRNA was measured by reverse transcriptase polymerase chain reaction( RT-PCR) method at 1,3,5,7,14 d after experiment in each group; and then the results were compared. Results There was no significant difference in SPA mRNA of lung tissues of newborn rats at 1,3 d after experiment in each group( P > 0. 05). The SPA mRNA level of lung tissues of newborn rats at 5,7 d after experiment in group 1 and group 2 was significantly lower than that in air control group( P < 0. 05). The SPA mRNA level of lung tissues of newborn rats at 5,7 d after experiment in group 1 and at 7 d in group 2 was significantly lower than that in group 3( P < 0. 05). The SPA mRNA level of lung tissues of newborn rats at 14 d after experiment in group 1 was significantly lower than that in air control group( P <0. 05). There was no significant difference in SPB mRNA of lung tissues of newborn rats at 1,3 d after experiment in each group( P > 0. 05). The SPB mRNA level of lung tissues of newborn rats at 5 d after experiment in group 1 and group 2 was sig-nificantly lower than that in air control group( P < 0. 05). The SPB mRNA level of lung tissues of newborn rats at 7,14 d after experiment in group 1 was significantly higher than that in air control group( P < 0. 05). The SPB mRNA level of lung tissues of newborn rats at 14 d after experiment in group 1 was significantly higher than that in group 3( P < 0. 05). There was no significant difference in SPC mRNA of lung tissues of newborn rats at 1,5 d after experiment in each group(
