摘 要:目的 :观察参芪保肾颗粒剂对阿霉素肾病大鼠转化生长因子 β1(TGF—β1)表达的影响 ,探讨本药延缓CRG进展的作用机理。方法 :采用阿霉素肾病大鼠 ,分为 5组 ,分别给予参芪保肾颗粒剂 2 0g/kg·d-1、5 0g/kg·d-1及 0 2 5 %苯那普利 10mg/kg·d-1灌胃 ,给药至第 18W。观察 2 4h尿蛋白定量、血肌酐、血尿素氮、肌酐清除率 ,肾小管—间质病理变化 ,并用免疫组化法观察转化生长因子 β1(TGF - β1)在肾组织的表达。结果 :给药组尿蛋白含量、血Scr、BUN明显降低 (P <0 0 1) ,肾小管—间质病理变化 ,给药组明显减轻 ,TGF—β1主要表达于肾小球内皮细胞、肾小管上皮细胞 ,给药组阳性表达率明显减少。结论 :参芪保肾颗粒剂可使尿蛋白量减少 ,降低血Ccr、BUN ,改善肾功能 ,减轻肾小管—间质损伤 ,使TGF—β1在肾脏组织内的表达明显下调 ,此可能是本药延缓CRF进展的作用机理之一。Objective:To observe the effect of shenbqibaoshen granule on the expression of TGF-β 1 in rats with adriamycin-induced nephrosis,discuss the mechanism of delaying the development of chronic renal failure.Methods:Adriamycin-induced nephrosis method was adopted,rats were divided into five groups.Quantitative test for uinary protein in 24hours,Scr,BUN and Ccr were detemined,renal tubulo-mesenchymal pathological changes were observed,expression of TGF-β 1 in renal tissue was observed by immunohistochemical method. Results:The urinary protein in 24 hours,Scr and BUN were obviously decreased ( P <0 01),the pathological changes of renal tubulo-mesenchymal was improved and expression of TGF-β 1 was decreased significantly in groups with medicines.Conclusion:Reducing urinary protein,blood Ccr and BUN,improving renal function and renal tubulo-mesenchymal injury,decreasing the expression of TGF-β 1 in renal tissue may be one of the mehanism of shenqihbaoshen granule to delay renal failure.
