重组水蛭素抗凝和抗血栓形成作用的实验研究

摘　　要：目的 :观察重组水蛭素对凝血系统、血栓形成和血小板聚集的影响 ,研究其药理作用。方法 :测定大鼠凝血时间 (CT)、活化部分凝血活酶时间 (APTT)、凝血酶时间 (TT)、凝血酶原时间 (PT)、纤维蛋白原含量 (Fib)及抗凝血酶Ⅲ (AT -Ⅲ )活性 ;采用A -V循环模型进行血栓形成实验 ;采用Born氏比浊法测定家兔血小板聚集。结果 :重组水蛭素可显著延长大鼠血浆的CT、APTT、TT及PT(P <0 0 0 1) ,明显降低Fib含量 (P <0 0 5 ) ,对AT -Ⅲ的活性无明显影响 ;明显减少血栓重量 (P <0 0 0 1) ;显著降低凝血酶诱导的家免血小板聚集率 (P <0 0 0 1) ,对胶原诱导的家兔血小板聚集率有降低趋势。结论 :重组水蛭素具有抗凝、抑制血小板聚集和血栓形成的作用。Objective:The anticoagulant activity and antithrombotic effect of recombinant (r-) hirudin made in China were studied in order to prove the experimental basis for the development of hirudin. Methods:Clotting time(CT)、activated partial thromboplastin time (APTT)、thrombin time(TT)、prothrombin time(PT)、fibrinogen level(Fib) and antithrombin Ⅲ(AT-Ⅲ) level in rats were determined.The method of carotid artery-external jugular vein by accessory pathway in rats was used,and platelet aggregation in rabbits were evaluated. Results: R-hirudin obviously prolonged CT、APTT、TT and PT and decreased Fib level in rat's plasma,but it had no effect on AT-Ⅲ.It also inhibited thrombus formulation in rat model of ateriovenous shunt thrombosis markedly,and inhibited thrombin-induced platelet aggregation obviously and collagen-induced platelet aggregation latently in rabbits.Conclusion:R- hirudin has marked properties of anticoagulant and antithrombotic action and preventing platelet aggregation as native hirudin.
