摘 要:使用RepeatMasker软件在栉孔扇贝的 6 935条ESTs中发现了 4 2条微卫星序列。选取其中的 7个序列 ,根据微卫星位点的旁侧序列设计引物 ,并对设计出的引物在中国、日本、韩国野生群体中进行多态性检测 ,发现有 6对引物能产生扩增产物 ,其中有 3对引物可望在以后的群体分析及连锁图谱的构建中加以应用。该结果初步证实了在栉孔扇贝的ESTs序列中存在微卫星位点 ,从而为扇贝生物中微卫星标记的筛选开辟了新的途径。Forty two microsatellite-containing sequences were identified from the 6935 EST sequences from scallop Chalmys farreri by using the on-line bioinformatics software of RepeatMasker. Based on the flanking sequences of the microsatellite loci, seven pairs of microsatellite primer were designed and applied to investigate the polymorphism in the wild populations of Chalmys farreri from China, Japan and Korea. Six pairs of primer generated PCR amplification products, and three of them yield polymorphic products that could be used in the study of population genetics and genetic mapping. The result indicated that there were microsatellite loci in the expressed sequences and ESTs analysis could provide a new approach to screen the microsatellite markers from scallop.
