摘  要:目的 研究乙酰胆碱(ACh)对内毒素血症时大鼠心肌肿瘤坏死因子-α(TNF-α)及氧自由基的影响。方法 雄性Wistar大鼠70只,分为7组。切断大鼠双侧颈部迷走神经干或做假手术对照,将左侧迷走神经干远端连接刺激电极,静注脂多糖(LPS,10 mg/kg体重)即刻持续电刺激(5 V,2 ms,1Hz)20 min,每隔 10 min重复刺激,每次 20 min,共刺激 3次。静注 LPS后 1.5 h检测各组心肌TNF-α、丙二醛(MDA)量及血ACh、肌酸激酶同功酶(CK-MB)、皮质醇水平。结果 迷走神经切断后电刺激复合静注 LPS组较单纯静注 LPS组血 ACh水平显著上升[(168.33±14.75)比(139.18±13.94)mg/L P<0.01],心肌组织中TNF-α、MDA量和血CK-MB水平显著降低[(103.52±14.65)比(147.24±33.88)ng/g pro;(.98±2.53)比(11.94±3.47)U/g;(1 135±304)比(1850±340)U/L(P均<0.01)]。结论 ACh可明显抑制LPS血症时大鼠心肌组织TNF-α和氧自由基生成,减轻心肌炎症反应,对心脏功能具有潜在保护作用。Objective To study the effects of acetylcholine on the tumor necrosis factor (TNF-α) and oxygen-derived free radicals in heart tissue in endotoxemic rats. Methods Seventy Wistar rats were randomly divided into 7 groups. Animals were subjected to bilateral cervical vagotomy, or a comparable sham surgical procedure in which the vagus nerves were isolated but not transected. The distal end of a vagus nerve trunk was placed across bipolar electrodes connected to a stimulation module and controlled by an acquisition system. Stimuli with constant voltage (5 V, 2 ms, 1 Hz) were applied to the nerve for 20 min immediately after administration of LPS (10 mg/kg, E coli O111: B4, Sigma), and then were repeat- ed 2 times after each of 10 min interval. Blood and tissue samples of these rats were collected at 1. 5 h af- ter LPS administration in all groups. TNF-α and MDA in heart tissue and serum corticosterone and acetylcholine were determined. Results The electrical stimulation of the efferent vagus nerve significant- ly decreased TNF and MDA contents in heart tissue and CK-MB in serum [ (103. 52±14. 65) vs (147. 24±33. 88) pg/mg pro, (7. 98±2. 53) vs (11. 94±3. 47) U/g, (1 135±304) vs (1 850±340) U/L(P < 0. 01)] and alleviated myocardial inflammation in heart tissue after endotoxin challenge. Con- clusion Acetylcholine can significantly alleviate the TNF-α, oxygen-derived free radicals in the heart tis- sue and myocardial inflammation during endotoxic challenged rats, thus it might produce a potential pro- tective effect on the heart during endotoxin in rats.
