摘 要:目的 对我国土拉弗菌进行基因分型研究。方法 应用短串联重复序列 (SSTR)分析技术对我国不同地区分离出的 5株土拉弗菌菌株进行分型。SSTR9和SSTR16两个区域扩增后测序 ,并将结果与国外其它分离株进行比较。结果 5株菌在SSTR9区重复序列的个数分别为 2 1、9、12、9、15 ,片段长度分别为 396bp、2 88bp、315bp、2 88bp和 342bp。在SSTR16区域扩增序列完全相同 ,拷贝数为 2 ,片段长度为 36 1bp。其中 4株菌重复序列多态性与瑞典、芬兰等欧洲株接近 ,而分离自新疆的114菌株在SSTR9区存在独特的重复序列多态性 ,可能是我国特有的基因型。结论 5个分离株均属于B型土拉弗菌 ,与型特异性引物分型结果一致。PCR SSTR分析可作为国内土拉弗菌分型及进行个体菌株区分的可靠方法。Objective The Francisella tularensis has been isolated from China for to investigate the genotyping of Francisella tularensis isolates from different areas of China. Methods The short sequence tandem repeat region (SSTR) analysis was used. The SSTR9 and SSTR16 regions in the five strains were amplified and sequenced. Results The size range and sequencing of the SSTR9 locus showed a high allelic variation. The repeat copy numbers of the five strains ranged from 9 to 21 and the amplicon sizes ranged from 288bp to 396bp. Four of them were similar with those isolated from Sweden, Finland and Norway, while one strain from Xinjiang (strain 114) was unique and its amplicon size was not reported in the previous study. On the other hand, SSTR16 locus in the five strains were identical. The copy number was 2 and the amplicon size was 361bp, which is in accordance with those observed in F.tularensis subsp. holarctica . Conclusion The five isolates investigated in the present study were all belong to the F.tularensis subsp. holarctica (type B). The analysis of short-sequence tandem repeats of F.tularensis strains appears to be a powerful tool for discrimination of individual strains in China.
