摘 要:目的 :建立压电石英晶体传感器检测血浆纤维蛋白原 (Fib)方法。方法 :采用AT切向、基频 10MHz的银膜石英晶体作为压电元件 :观察血浆凝集过程中晶体振荡频率的变化 ,确定血浆凝集反应的起点和终点。建立“浓度 -时间”标准曲线 ,并检测 6 4个临床标本。结果 :( 1)纤维蛋白原试剂加入反应体系后 ,晶体振荡频率变化呈前后两点凝集峰再缓慢降低达到平稳 ,计算两点之间的时间即为血浆凝集时间。“浓度 -时间”标准曲线线性关系良好。 ( 2 )该方法的检测结果与STAGO磁珠法的相关性好 ,γ =0 91。结论 :压电石英晶体传感器检测血浆纤维蛋白原 ,血浆凝集反应起点和终点确定方便、可靠 ,且操作简单、快速 ,成本低廉 ,检测结果准确 ,是一种敏感性高、重复性好的血浆纤维蛋白原检测新方法 ,可用于常规检测。Objective: To establish a piezoelectric quartz crystal sensor for determination of the fibrinogen of plasma. Methods: Silver coated quartz crystal by 10MHz quartz AT cut was used as electrodes. The oscillating frequency changes were observed in the course of the coagulation reaction of plasma sample to assure the start and end point of the coagulation reaction to establish “concentration coagulation time” criteria curve. Results: After fibrinogen reagent was added into the reaction system, oscillation frequency of the crystal was decreased as a ladder, then rose abruptly and descended quickly to balance. The point of the ladder like frequency beginning to decrease was regarded as the start point of coagulation reaction and the turning point of the frequency from rise to decrease as the end point. The time between two peaks was calculated as the whole time of plasma coagulation. It suggested that there should be a significant correlation between the results from this method and STAGO method (r=0.91). Conclusion: This method for fibrinogen detecting is simple, rapid, specific, accurate and cheap, which might be a valuable method for detection of fibrinogen in clinical laboratory.
