摘 要:目的 :观察糖尿病大鼠大脑中动脉病变特点及氯沙坦对其的影响。方法 :健康雄性 Wistar大鼠 2 3只 ,随机分为正常对照组和糖尿病组 ,经链脲佐菌素诱发糖尿病大鼠模型。再将糖尿病组随机分为糖尿病对照组 (DM1组 ) ,糖尿病氯沙坦治疗组(DM2组 )。DM2组接受氯沙坦 1 0 m g/ (kg· d)灌胃给药 ,1 4周后处死。采用放射免疫法检测血浆内皮素、血管紧张素 含量 ,观察大鼠大脑中动脉病理学变化以及超微结构。结果 :DM1组血浆内皮素 [(4 88.4± 90 .4 ) ng/ L]高于正常对照组 [(2 6 0 .9±2 7.7) ng/ L]及 DM2组 [(36 6 .0± 4 8.7) ng/ L],DM1组血管紧张素 [(1 1 0 8.6± 6 3.1 ) ng/ L]高于正常对照组 [(395 .6±4 5 .4 ) ng/ L],但低于 DM2组 [(1 36 6 .4± 5 7.7) ng/ L](均 P <0 .0 5 )。DM1组大脑中动脉呈动脉硬化性改变 :平滑肌增殖明显 ,动脉壁厚度、动脉管壁面积均明显高于 NC及 DM2组 (P <0 .0 5 )。电镜观察该组大脑中动脉病变明显重于正常对照组、DM2组 :内皮细胞变性肿胀 ,部分坏死脱落 ;平滑肌肥大排列紊乱 ,溶解坏死 ,间隙增宽 ,电子致密物增加。DM2组内皮细胞变性较轻 ,无坏死脱落 ,平滑肌增殖被抑制 ,管壁增厚及管腔狭窄均较轻。结论 :糖尿病大鼠大脑中动脉内皮细胞损伤 ,功能障碍 ,平滑肌增?Objective:The aim of this study was to demonstrate histopathological alternation of the middle cerebral arteries in streptozotocin induced diabetic rats and assess the possible effect of losartan on the alternation.Methods:Male wistar rats were randomly divided into normal control group (NC,n=8),diabetic control group(DM1,n=7)and diabetic group treated with losartan(DM2,n=8)10 mg/(kg·d). Diabetic rats were streptozotocin induced. Sham injection, control rats were studied in parallel. Three groups were administered for 14 weeks after injection. In the 14th week, plasma levels of edothelin(ET), angiotensinII (AngⅡ) were determined by radioimmunoassay respectively. The arteries were examined under light and electron microscope.Result:In DM1 group, plasma levels of ET and AngⅡ were increased significantly compared with NC group(P<0 05).In DM2 group, plasma level of AngⅡ were elevated more significantly while plasma ET level declined (P<0 05) compared with DM1 group. There were mitochondria edema vacuolization and some endothelial cells necrosis and exfoliation. The multiplication of smooth muscular cells in the artery wall was prominent The artery wall, in DM1 group ,became thicker than that in NC group, but a little pathological changes in DM2 group.Conclusion:Plasma levels of ET and AngⅡof diabetic rats are increased. The structure and function of the MCAs undergo significant alternation in DM1 group. Losartan can effectively prevent the alternation.
