广西地区壮、汉族人群载脂蛋白B基因多态性及其与冠心病的关系

摘　　要：目的 :探讨载脂蛋白 B(Apo B)基因的多态性在广西地区壮、汉两族人群中的分布特点及其与冠心病发生的关系。方法 :应用聚合酶链反应 (PCR)对 1 1 1例健康人 (正常组、对照组 ,壮族 6 2例、汉族 4 9例 )和 5 9例冠心病 (CHD)患者 (冠心病组 ,壮族 1 7例 ,汉族 4 2例 )的 apo B基因的 Xba I、Eco RI位点限制性片段长度多态性 (RFL P)以及 3′端可变数串联重复序列 (3′VNTR)多态性进行研究。结果 :1两族正常组与对照组在 Xba I的等位基因分布没有区别 ;2 3′ VNTR等位基因的分布各有特点。广西壮族人群以 HVE30、HVE32为主 ,广西汉族人群以 HVE34、HVE36和 HVE38为主 ,而 CHD组两个民族均以HVE32、HVE34为主 ;3Eco RI位点等位基因和 3′ VNTR大于或等于 38的大等位基因的分布频率在广西汉族两组人群之间的差异有统计学意义 (P <0 .0 5 )。结论 :广西汉族人群冠心病的发病与 Apo B基因的 3′ VNTR及 EcoObjective:To study the polymorphisms of the apoliprotein B(ApoB) gene difference between the Zhuang nationality and Han nationality of Guangxi population,and the association between the polymorphisms of the apoliprotein B gene and the coronary heart disease(CHD).Methods:The restriction fragment length polymorphisms of XbaI and EcoRI sites and the 3′VNTR of ApoB gene in 111 healthy people(Zhuang 62,Han 49),59 CHD(Zhuang 17,Han 42) in Guangxi region were studied by Polymerase Chain Reaction(PCR) Result:There was no significal differences between the control and CHD in Zhuang of XbaI and EcoRI allelic distribution.But the E - frequency was higher in Guangxi Han CHD than in the control. The 3′ VNTR allelic distributions in the two nationality groups displayed their characters respectively. There were mainly HVE30?HVE32 alleles in Guangxi Zhuang healthy population; there were mainly HVE34,HVE36 and HVE38 allels in Guangxi Han healthy population,and there were mainly HVE32 and HVE34 allels in CHD of Zhuang and Han.The frequency of 3′ VNTR allele (number of repeated units≥38) was higher in Guangxi Han control than in CHD.Conclusion:The 3′ VNTR alleles and EccoRI site are link to the CHD in Guangxi Han.
