摘  要:目的 研究NF κB在多器官功能不全小鼠各脏器中的活性变化 ,以期探讨其在多器官功能不全发生过程中的作用。方法 利用酵母多糖制作多器官功能不全综合征的小鼠模型 ,提取小鼠脏器的细胞核蛋白 ,用凝胶阻滞分析方法观察NF κB的活性变化。结果 酵母多糖腹腔注射后 ,小鼠发生了三个阶段的生理学变化 ,表现为体温和体重的波动 ;在第三阶段小鼠脏器发生了严重的病理学改变。同时 ,NF κB的DNA结合活性有显著性增加。结论 酵母多糖有效地诱发了小鼠发生多器官功能不全综合征。在此模型小鼠的各脏器中 ,NF κB活性显著性增加 ,提示NF κB可能是多功能不全发生过程中的一个关键的转录因子。Objective Nuclear factor κB (NF-κB) is a ubiquitous transcription factor in mammalian cells. It was reported that NF-κB regulated the expression of genes responsible for multiple biological processes including immune responses, inflammatory reaction, cell proliferation and apoptosis. The present study was to examine the changes in activity of NF-κB in the mouse organ with multiple organ dysfunction syndrome(MODS) in order to study the function of NF-κB in the development of this syndrome. Methods Mouse model for MODS was formed by zymosan intraperitoneal injection. Nuclear extracts from the organs of MODS model were examined by electrophoretic mobility shift assays (EMSA) to assay the activity of NF-κB. Results Three phases of physiological changes developed after zymosan intraperitoneal injection and were characterized by the changes in body weight and temperature. Mouse organs developed severe inflammation and the activity of NF-κB increased remarkably. Conclusion Mouse model for MODS was formed successfully by zymosan treatment and NF-κB activity increased in this model. The results suggest that NF-κB play an important role in the development of MODS.
