摘 要:以无根石竹试管苗为试材,通过外施多胺及多胺生物合成抑制剂调控石竹(DianthuschinensisL.)试管苗成花,研究成花过程中石竹内源激素含量的变化。结果表明,外源腐胺(Put)、亚精胺(Spd)、精胺(Spm)及多胺生物合成抑制剂二氟甲基鸟氨酸(DFMO)、二氟甲基精氨酸(DFMA)、环己胺硫酸盐(CHAS)能调控石竹试管苗成花率;Spm、Spd分别将成花率由38.3%提高至48.3%和60.0%,也提高了内源玉米素核苷类(ZRs)、异戊烯腺嘌呤类(iPAs)、吲哚 3 乙酸(IAA)水平;DFMO、CHAS则完全抑制成花,显著降低内源IAA水平。因此,内源激素与多胺的代谢水平共同控制石竹试管苗成花。Small pieces of young leaves of Dianthus chinensis L.collected from the campus of Nanjing Forestry University in spring were cultured on the differentiation medium of MS+6BA(2.0 mg/L)+2,4D(0.2 mg/L) after sterilization.Green yellow calli were induced from the explants after one week of culture and then were subcultured on the differentiation medium of MS+KT(0.3 mg/L)+6BA(0.3 mg/L)+NAA(0.1 mg/L).Four weeks later,shoots appeared in some culture bottles.4~5 cm long shoots were cut at the bottom and the same amount of shoots(60) were cultured on control MS medium and treat MS media supplemented with 0.05 mmol/L putresine(Put),spermidine(Spd),spermine(Spm)and 5 mmol/L difluoromethylornithine(DFMO),difluoromethylarginine(DFMA),cyclohexyl ammolonium sulphat(CHAS)respectively.The experiments were repeated 4 times.Flower buds appeared on some media two weeks after inoculation and then flowered 4~5 days later.The apexes were collected every 5 days from the beginning day of culture for detecting the contents of zeatin tiboside group (ZRs),isopentenyl adenine group(iPAs) and indole3acetic acid(IAA).The culture results showed that Spd and Spm promoted flowering significantly,and the flowering rate was raised from the control of 38.3% to 48.3% and 60.0% respectively.Put and DFMA didn't affect flowering much while DFMO and CHAS inhibited flowering totally.Endogenous hormone content detection results showed that Spd and Spm enhanced the contents of ZRs,iPAs and IAA while DFMO decreased them notably,and Put didn't affect them much.CHAS and DFMA enhanced the contents of ZRs and iPAs while decreasing the contents of IAA,and the effects of DFMA were not as notable as that of CHAS.It is concluded that exogenous PAs and PA biosynthetic inhibitors can affect the flowering and metabolism of hormones of D.chinensis L.Spd and Spm promote flowering and the contents of ZRs,iPAs and IAA while DFMO and CHAS inhibit flowering and decrease the contents of IAA,which indicate that ZRs,iPAs and IAA are re
