摘 要:采用直接比色法测定板蓝根中靛蓝、靛玉红的质量分数。对靛蓝、靛玉红在 2 4 0~ 70 0nm波段进行扫描 ,确定其最大吸收波长分别为 6 0 1和 5 31nm。分别通过单因素试验和正交试验 ,确定提取靛蓝、靛玉红的最佳工艺参数。结果表明 ,乙醇体积分数为 75 %时 ,靛蓝和靛玉红的提取物得率最高 ;提取时间为 4h时 ,靛蓝和靛玉红的提取物得率最高。正交试验确定的最佳提取工艺参数为 :药材粒度 4 0目 ,提取时间 4h ,乙醇体积分数 75 % ,此时 ,提取物得率达 1 74 %。Extraction and determination of mass ratio of Indigo and Indirubin in Indigowoad root were studied. Indigo and Indirubin were scanned from 240 to 700*!nm. From the spectrogram we can see that the largest absorbing wavelength of Indigo and Indirubin was 601 and 531*!nm respectively. The best parameters of technology have been determined through single factor and orthogonal test on extracting indigo and Indirubin respectively, results showed that ethanol with a volume ratio of 75% was suitable solvent to extract Indigo and Indirubin and 4 hours was the proper time to extract Indigo and Indirubin. Through orthogonal test the best parameters of technology were obtained. The results are as the following: the herbal granularity is 40 eye, extraction time is 4*!h, ethanol with a volume ratio of 75%. On this condition, the extracting rate was up to 1.74%.
