当归挥发油对子宫的双向作用及其活性部位筛选

摘　　要：目的　研究当归挥发油 (AN)及其酸性部位 (A1)、酚性部位 (A2 )和中性非酚性部位 (A3)对子宫的作用并筛选出具有最佳抑制作用的活性部位。方法　采用离体子宫肌张力记录法 ,薄层色谱和气相色谱 质谱分析法。结果　①当归挥发油对正常离体大鼠子宫收缩功能呈双向作用 ,小剂量 (≤ 2 0mg/L)略有兴奋作用 ,大剂量 (≥160mg/L)则明显抑制之。较大剂量AN可浓度依赖性地抑制缩宫素诱发的子宫兴奋作用 ,也可明显抑制高钾引起的子宫收缩。②色谱分析结果表明A3 活性部位的组成明显不同于挥发油、A1和A2 。在正常离体大鼠子宫标本 ,A1(0～160mg/L)呈兴奋作用 ,仅在 3 2 0mg/L才出现明显的抑制作用 ;小剂量A2 (≤ 10mg/L)略有兴奋 ,而大剂量 (≥ 2 0mg/L)表现为抑制作用 ;A3 (10～ 160mg/L)均表现为抑制作用 ,其IC5 0 为 3 2 5mg/L ,抑制效价强度是挥发油的 3 7倍。结论　当归挥发油对子宫收缩功能的影响 ,依剂量呈现小剂量兴奋、大剂量抑制的双向性特征 ;A3Objective To study the effects of Angelica na phtha (AN) on rat uteruses and screen out the most effective component on the in hibition of the myometrium contraction. Methods The smooth muscle tension of the uteruses was recorded. Active components of AN were identified by TLC and GC-MS/MSD. Results AN had dualistic effects on the contraction of normal i solated uteruses. The uteruses could be slightly excited by low concentration ( ≤20 mg/L) and distinctly inhibited by high concentration (≥160 mg/L) of AN. Th e excitement induced by oxytocin could be concentration-dependently inhibited b y AN. Three active components (A 1, A 2, A 3) were extracted from AN. A 1 ( ≤160 mg/L) could enhance the contractile activity of normal isolated uteruses e xcept at the concentration of 320 mg/L. A 2 (≤10 mg/L) had weak exciting effec ts while it (≥20 mg/L) had inhibitory effects. All concentrations of A 3 (10— 160 mg/L) could decrease the contractile force, with IC 50 value of 32 5 m g/L. Moreover, the inhibitory potency of A 3 was 3 7 times as strong as that o f AN. Conclusion The concentration-effect curve of AN on isola ted uteruses possesses dualistic property. Among the three components extracted from AN, A 3 is the most effective one.
