摘 要:目的:观察β-七叶皂甙钠(SodiumAescinate,SA)对脊髓继发性损伤的保护作用。方法:参考Nystr?m方法建立大鼠脊髓压迫伤模型(T8~9),将动物随机数字表法分为假手术组、模型组、SA组、甲泼尼龙(MP)组。应用免疫组化和图像定量分析法观察脊髓损伤后天冬氨酸特异性半胱氨酸蛋白酶(caspase)-3蛋白的表达,测定不同药物组超氧化物歧化酶(SOD)活性和丙二醛含量的变化。术后1,2周联合行为学评分(CBS)观察实验动物神经功能恢复情况。结果:假手术组有少量散在caspase-3蛋白的表达,脊髓损伤模型组cas-pase-3蛋白阳性细胞增多,SA组和MP组阳性细胞数均低于模型组,两治疗组间比较无显著性差异。SA组脊髓组织丙二醛含量明显低于各时相点模型组,SOD活性显著增高,与MP组比无显著性差异。CBS评分SA组明显增加。结论:SA可以下调caspase-3蛋白的表达,抑制细胞凋亡,缓解脊髓损伤后的脂质过氧化,从而起到脊髓保护作用。AIM:To investigate the protective effects of sodium aescinate(SA) on experimentally secondary injured spinal cord in rats. METHODS:A compression injured model(T8 9) of rat spinal cord was established according to Nystrm.Rats were randomly divided into four groups:sham operation group, spinal cord injury(SCI)group,SCI with SA treatment group,and SCI with methylprednisolon(MP) treatment group.The caspase 3 protein was evaluated with immunohistochemical and image quantitative analysis.The superoxide dismutase(SOD)activity and malondialdehyde(MDA)content of the injured spinal cord were measured after reagents were injected intraperitoneally.Functional status of nerve was assessed by the modified combined behavioral score(CBS)at 1 and 2 weeks after operation respectively. RESULTS:Little expression of caspase 3 protein was found in sham opera tion group.In SCI group,expression of caspase 3 protein was increased.Compared to the SCI group,less expression of caspase 3 protein was noted in the SA and MP treatment groups,and no significant difference was found between the two treatment groups.In the SA treatment group, the SOD activity was increased and the level of MDA was reduced markedly(P< 0.01),which were no significant difference in the MP treatment group,and CBS was elevated significantly as well. CONCLUSION:SA can downregulate the expression of caspase 3 protein, prevent apoptosis and alleviate the lipid peroxidation.It may play a protective role in early SCI.
