摘 要:本文用程序外DNA合成(UDS)试验方法检测氟、砷及氟砷联合染毒对大鼠肝脏DNA的损伤及修复的影响。将雄性Wistar大鼠随机分为四组:氟组、砷组,氟砷联合组及对照组。分别饮用含氟化钠125ppm、三氧化二砷75ppm、氟化钠125ppm加三氧化二砷75ppm的水及蒸馏水。结果染毒3个月的大鼠肝脏,在氟组其cPm均值为1580.7±196,明显高于对照组377±34。砷组的cPm均值为239.3±104,低于对照组。氟砷联合组的cPm均值为309.3±53,接近于但略低于对照组。染毒6个月脾大鼠肝脏,氟、砷及氟砷联合组的cPm均值均低于对照组,其掺入百分率分别为对照组的48.3%(氟组)、38%(砷组)、87%(氟砷联合组)。此结果说明慢性氟、砷及氟砷联合中毒可以引起大鼠肝细胞的DNA损伤,并影响其修复损伤的能力。另外,本实验的结果还表明氟与砷有拮抗作用。Unscheduled DNA synthesis was measured by scintillation counter in primary cultures of rat hepatocytes.Rats were fed with sodium fluoride(Na F),trivalent arsenic trioxide(As2O3)and NaF combined with As_2O_3,respectively,indrinking water. Treatment with the NaF for three months resultec in the induction of UDS.The average value of  ̄3H thymidine incorporation in NaF group was 1580.7±197cpm,It was significantly greater than that in control group(377±34cpm).In As2O3 group the average value(239.3±104cpm)was lower than that in control group.However.in combined NaF with As_2O_3 group the cpm value (309.3±53)was closed to that in control group .Exposure of rats to the chemicals for six months caused a lower ̄3H thymidine incorporation in all three treated groups than that in control group.The percentage of cpm in NaF,As2O3 and combined group was 48.3%,38% and 87% in turn.These results indicated that 6-month exposure of rats to NaF,As_2O_3 or combined NaF with As_2O_3 could cause DNA damage and inhibit DNA repair.In addition,these results suggested that fluoride and arsenic could be antagonist.
