摘　　要：目的　构建 MC1 4 8腺病毒重组体。方法　从传染性软疣病毒基因组中克隆 MC1 4 8基因 ,产物测序后将之插入柯斯质粒 ,然后与 DNA- TPC共转染进 2 93细胞 ,同源重组构建腺病毒重组体。浓度梯度法测定病毒滴度。结果　成功构建 Ad- MC1 4 8腺病毒重组体 ,经测定滴度达 3.5 6× 1 0 9(pfu/ml)。结论　本方法建立为研究 MC1 4Objective\ To construct recombinant adenovirus Ad MC148.Methods\ MC148 gene was cloned from molluscum contagiosum virus(MCV)and inserted into cosmid vector,which was then cotransfected with DNA TPC into 293 cells to construct recombinant Ad MC148.The titer of recombinant adenovirus was detected by series dilution.Results\ Recombinant Ad MC148 was constructed and its titer was 3 56×10 9(pfu/ml).Conclusion\ The method is the base for further study of MC148 gene antagonizing chemokines.
